Increased plasma levels of platelet-derived growth factor (PDGF-BB + PDGF-AB) in patients with never-treated mild essential hypertension

Abstract: Platelet-derived growth factor (PDGF) could play a role in both vascular hypertrophy and atherosclerotic disease associated with hypertension. To assess whether plasma PDGF level is increased in mild essential hypertension, we measured plasma PDGF concentration in 25 never-treated patients with uncomplicated mild essential hypertension and in 22 normotensive healthy subjects. To evaluate the contribution of platelets to plasma PDGF in the two groups, we also measured plasma beta-thromboglobulin (BTG). Measurem… Show more

“…Peripheral blood smears were also evaluated. Previous reported data for collecting samples of another mitogenic factor that is also stored in α-granules of platelets, platelet-derived growth factor (PDGF), was used in order to collect PPP and avoid ex vivo platelet activation [17, 18, 19, 20]. 4.5 ml of blood were mixed in pre-chilled plastic tubes containing 0.5 ml of an anticoagulant consisting of 15 mmol/l theophylline, 3.7 mmol/l adenosine, 0.198 mmol/l dipyridamole and 0.11 mol/l sodium citrate.…”

“…Increased PDGF-BB levels play an important role in the vascular structural changes associated with hypertension (57). Our data showed that PDGF-BB levels increased gradually as the severity of the hypertension disorders increases.…”

“…Platelets, macrophages, and endothelial cells can produce both PDGF chains (A and B), whereas vascular SMC produce only PDGF-A chains. 36 As we only measured PDGF-BB, this did not represent vascular SMC-produced PDGF and may have only reflected platelet, macrophage, or endothelial-released PDGF. However, PDGF-BB is known to be a potent SMC stimulator.…”
