Increased osteoinductivity and mineralization by minimal concentration of bone morphogenetic protein-2 loaded onto biphasic calcium phosphate in a rabbit sinus

Abstract: PurposeThe purpose of the present study was to evaluate the effectiveness of a minimal concentration of bone morphogenetic protein-2 (BMP-2) in terms of quantitative and qualitative analyses of newly formed bone in a rabbit maxillary sinus model.MethodsIn 7 rabbits, sinus windows were prepared bilaterally. Biphasic calcium phosphate (BCP) loaded with 0.05 mg/mL BMP-2 was grafted into one sinus (the BMP group) and saline-soaked BCP was placed into the other (the control group) in each animal. The animals were a… Show more

“…), ACS (Choi, Lee, Kim, Kim & Choi, ) and BCP (Choi et al., ; Kim, Cha, et al. ; Kim, Cha, Lee, Choi & Cho, ). Moreover, bMSCs modified with AdBMP‐2 and combined with different scaffolds promoted significantly more new bone formation in the rabbit maxillary sinus compared to sites where AdBMP‐2 was not employed (Jiang, Sun, Lai, Zhao & Wang, ; Sun, Xia, Chou, Zhong & Zhang, ).…”

“…However, this study spanned only eight weeks, so more research is needed to investigate how bone derived by grafting changes over time. It will also be necessary to examine concentration-dependent bone formation by BMP-2 in order to find the most effective dose [8,26]. Manipulating the composition of BC could generate new material that is even safer and more effective in the long term.…”

“…A stock human-derived recombinant BMP-2 (rhBMP-2) solution (1.0 µg/µL) (Osteogenetics GmbH, Würzburg, Germany) was diluted to 0.5 μg/μL with sterilized distilled water (Otsuka distilled water, Otsuka Pharmaceutical, Tokyo, Japan) [26].…”

“…Regarding the dosage of BMPs, the combination of BCP with a high concentration of rhBMP-2 (1.5 mg/ml) has proved to inhibit bone regeneration from pristine bone and increase the inflammatory response in the early stages in animal models [169] . Meanwhile, a low dosage of rhBMP-2 (0.05 mg/ml) in a BCP carrier has been shown to promote an osteoinductive effect with accelerated mineralization in animal models [170] . Moreover, it has been reported that a combination of BCP with 0.5 mg/ml of rhBMP-2 formed a greater volume of bone in a rabbit model than BCP combined with 1.0 mg/ml of rhBMP-2 [171] .…”
