Increased Na+/H+ antiport activity in the renal brush border membrane of SHR

Abstract: Defect in renal salt excretion may play an important role in the pathogenesis of hypertension. We examined sodium (Na+) uptake by brush border membrane (BBM) vesicles of young (6 week old) spontaneously hypertensive rats (SHR) and normotensive Wistar-Kyoto rats (WKY) of the same age. SHR had lower urinary Na+ excretion (223.1 +/- 9.3 vs. 266.3 +/- 3.7 microEq/day/100 g, N = 8, P less than 0.01) and higher systolic blood pressure (98.9 +/- 1.2 vs. 82.9 +/- 1.8 mm Hg, N = 8, P less than 0.01) than WKY. BBM vesic… Show more

“…Compared with proximal tubules from WKY rats, there was increased NHE3 activity in tubules from SHR. These findings are compatible with previous results; Morduchowicz et al, 1989;Dagher and Sauterey, 1992;Hayashi et al, 1997). Furthermore, we showed that the sensitivity of the enhanced NHE3 activity in proximal tubules was about 50-fold higher in SHR compared with WKY rats.…”

“…22 Na uptake studies were performed according to Morduchowicz et al (1989). In brief, purified BBMV were prepared by Mg 2ϩ precipitation method from SHR and WKY treated with Y-27632 or vehicle in vivo.…”

“…Furthermore, nifedipine consistently elevated intracellular pH values (23). An enhanced activity of the H + -Na + antiporter (as a major mechanism of cell defense against cellular acidification) has been reported in lymphocytes (24) as well as in the renal brush border membrane of spontaneously hypertensive rats (25) and in hypertensive rats (26). Other authors have shown enhanced responsiveness of the renal proximal Na + -H + antiporter of hypertensive rats to stimulation with some hormones (27).…”

“…This enhanced responsiveness in SHR kidney may be partially explained by α 2B -adrenoceptor-mediated acceleration of Na + /H + exchange. The latter may be explained by an increased activity of the Na + /H + exchanger (NHE) in the kidney of the hypertensive rat strains, namely the SHR (44,45) that is unresponsive to inhibitory action (46). This increased expression of NHE in the kidney of SHR compared to WKY rats (47) has been related to the development of hypertension in these animals (48).…”
