Mast cells participate in allergies and inflammation by secreting a variety of pro-inflammatory mediators. Curcumin, the active component of turmeric, is a polyphenolic phytochemical with anti-tumor, anti-inflammatory, anti-oxidative, and anti-allergic properties. The effects of curcumin on compound 48/80-induced mast cell activation and passive cutaneous anaphylactoid reactions are unknown. In this report, we investigated the influences of curcumin on the passive cutaneous anaphylactoid response in vivo and compound 48/80-induced mast cell activation in vitro. The mechanism of action was examined by calcium uptake measurements and cAMP assays in mast cells. Curcumin significantly attenuated the mast cell-mediated passive cutaneous anaphylactoid reaction in an animal model. In agreement with this in vivo activity, curcumin suppressed compound 48/80-induced rat peritoneal mast cell (RPMC) degranulation and histamine release from RPMCs. Moreover, compound 48/80-elicited calcium uptake into RPMCs was reduced in a dose-dependent manner by curcumin. Furthermore, curcumin increased the level of intracellular cAMP and significantly inhibited the compound 48/80-induced reduction of cAMP in RPMCs. These results corroborate the finding that curcumin may have anti-allergic activity.

Several studies have demonstrated the potential of flavonoids for the prevention and/or intervention of various allergic diseases. One such naturally occurring flavonoid is silibinin ( Fig. 1), which is the major biologically active constituent in silymarin that is a defined mixture of polyphenolic flavonoids isolated from the milk thistle (Silybum marianum) extract. Silibinin has high human acceptance being used clinically and consumed as dietary supplement around the world for its strong anti-hepatotoxic efficacy.1,2) A wide range of studies have shown that silibinin exhibits cancer-preventive and anti-carcinogenic effects.3,4) Moreover, silibinin possesses a variety of additional biological effects, such as antioxidative 5) and anti-inflammatory effects. 6)Immediate-type hypersensitivity (anaphylaxis) is an acute and life-threatening allergic reaction that occurs massively and systemically. Mast cells play a key role in allergic diseases through the release of a number of mediators and cytokines.7) The secretory response of mast cells can be induced by aggregation of their cell surface-specific receptors (FceRI) for immunoglobulin (Ig) E by the corresponding antigen.8) Activated mast cells can produce histamine, as well as a wide variety of other inflammatory mediators such as leukotrienes, prostaglandins, proteases and several pro-inflammatory and chemotactic cytokines such as tumor necrosis factor (TNF)-a and interleukins (IL)-6, IL-4, IL-13, IL-8.9) Among these substances, histamine remains the bestcharacterized and most potent vasoactive mediator implicated in the acute phase of immediate hypersensitivity. 10)Mast cell degranulation can also be elicited by non-immunologic stimulators such as neuropeptides, basic compounds, complement components and certain drugs.11) Compound 48/80, a mixed polymer of phenethylamine cross-linked by formaldehyde, is the potent secretagogue for mast cells.12) It vigorously activates cellular exocytosis, inducing a rapid release of inflammatory mediators including such as histamine. Thus, compound 48/80 has been used as a direct and convenient reagent to study the mechanism of allergy and anaphylaxis.13) Additionally, murine mast cell is a good experimental model for the study of the mechanism of stimulus-secretion coupling.In the earlier work, Lecomte and Damas reported mast cell protection against dextran induced by silybin (silibinin). 14)Moreover, it was shown that silybin inhibited anaphylactic shock in rats sensitized by ovalbumin and concluded that silybin seemed to be a stabilizer of mast cell membrane, thus preventing histamine release. 15) Furthermore, Miadonna et al. demonstrated that silybin, one of the pure active agents in silymarin, might have anti-allergic activity via inhibition of histamine release from human basophils.16) Recently, silymarin has been suggested to have a protective effect in the early-phase of allergic asthma.17) Nevertheless, to our knowledge, a detail relationship between silibinin and mast cellmediated anaphylaxis-like reactions has not y...

Mast cells are important mediators of immediate hypersensitivity, such as allergy and anaphylaxis. In response to the IgE-dependent and IgE-independent stimuli, mast cells elicit degranulation, which results in releasing various mediators, such as histamine and an array of inflammatory cytokines. Histamine, a main content of granules in mast cells, exerts many effects related to the acute phase of immediate hypersensitivity, including vasodilatation, increased vascular permeability, and tissue edema. 1) Mast cell degranulation can also be elicited by the basic secretagogues. The most potent secretagogues include the compound 48/80, which is a mixed polymer of phenethylamine cross-linked by formaldehyde. 2)In general, compound 48/80 initiates the generation of superoxide anion by A-kinase inactivation through decreasing the intracellular cAMP concentration in mast cells.3) Generated superoxide anion results in the increase of intracellular calcium content, which leads to histamine release from mast cells.4) Thus, compound 48/80 has been used as a direct and convenient reagent to study the mechanism of allergy and anaphylaxis. 1)Phellinus linteus, an orange color mushroom growing well on mulberry tree, is a well-known fungus of the genus Phellinus in the family of Hymenochaetaceae and has been used as a traditional herb medicine in oriental countries. 5)Proteoglycan isolated from Phellinus linteus has been shown to have anti-tumor and immunomodulatory effects.6) It has also been shown to alleviate the septic shock induced by the lipopolysaccharide injection in mice and to be involved in the prevention and treatment of autoimmune joint inflammation such as rheumatoid arthritis. 7,8) Moreover, ethanol extract of Phellinus linteus exhibits potent anti-oxidant and anti-angiogenic activities.9) Lim et al. have reported that organic solvent extracts of Phellinus linteus suppress IgE production by modulating helper T lymphocytes 1/2 balance in murine splenocytes and mesenteric lymph node lymphocytes. 10,11) Inagaki et al. have also demonstrated that a boiling water fraction from mycelium of Phellinus linteus inhibits IgE-dependent mouse triphasic cutaneous reaction.12) However, direct cellular mechanism underlying the mast cell-mediated anaphylaxis-like reaction has poorly been understood. In this study, we have investigated the inhibitory mechanism of water extract from the fruiting body of Phellinus linteus against the anaphylaxis-like reaction and mast cell activation. MATERIALS AND METHODS MaterialsCompound 48/80, disodium cromoglycate (DSCG), anti-DNP IgE, DNP-human serum albumin (HSA), bovine serum albumin (BSA) and HEPES were purchased from Sigma Chemical Co.
