Increased Glucosinolate Production in Brassica oleracea var. italica Cell Cultures Due to Coronatine Activated Genes Involved in Glucosinolate Biosynthesis

Abstract: In this work, the effect of different elicitors and culture conditions on the production of glucosinolates in broccoli cell cultures was studied. The results showed that 0.5 μM coronatine was the best elicitor for increasing glucosinolate production (205-fold increase over untreated cells after 72 h of treatment). Furthermore, the expression levels of some genes related to the biosynthetic pathway of glucosinolates as well as three Myb transcription factors also have been studied. The highest glucosinolate lev… Show more

“…The use of PHEN and DPI applied individually caused a high repression in the expression of CYP79B2 gene, suggesting that JA and ROS play a key role in the wound-induced activation of indolic glucosinolate biosynthesis, whereas ET plays a slight role. These observations agree with previous findings in Brassica species [15,19,20]. For instance, in Arabidopsis, CYP79B2 was reported to be inducible by wounding and by MeJA treat-ment [15].…”

“…For instance, in Arabidopsis, CYP79B2 was reported to be inducible by wounding and by MeJA treat-ment [15]. Likewise, in broccoli cell culture, CYP79B2 was reported to be highly induced by coronatine treatment (mimicking JA action) and was closely correlated with an increase in GS content [19]. Furthermore, 1-MCP application during postharvest of broccoli was reported to reduce CYP79B2 gene expression during storage [20].…”

“…This induced expression pattern of GSL genes was also consistent with the total GSL content in transgenic lines, which implies that BrPP5.2 can regulate GSL content by modulating the GSL biosynthesis-related genes through yet-to-be-identified mechanisms. Of the upregulated DEGs, the upregulation of MYB28 [ 38 ]; CYP83B1 , MYB122 , and ST5a [ 39 ]; ST5b and ST5c [ 40 ]; MAM3 [ 41 ]; and MAM1 and CYP83A1 [ 42 ] was shown to increase the GSL content. Other genes such as CYP79F1 , BCAT4 , AOP2, UGT74C1, BAT5 [ 43 ], and sulfotransferases ( ST5b , ST5c ) [ 44 ] have been shown to participate in various steps of GSL biosynthesis and its modification.…”

“…We also identified and quantified individual GLSs from the whole edible parts of Chinese kale plants demonstrating that the content of all GLSs was significantly enhanced by liquiritin treatment at 500 and 750 ppm (Table ). Similar types of GLSs have been previously identified in Chinese kale by Sánchez‐Pujante et al ., Cuong et al . and Wang et al…”

“…Changes in the expression levels of various genes involved in GLS and PH biosynthesis were assessed, according to previous reports . For that purpose, another independent experiment was performed in the greenhouse, including the same treatments and culture conditions described above.…”
