Increased expression of von Willebrand factor gene is associated with poorer survival in primary lower grade glioma

Abstract: Background: Venous thromboembolism is a common complication in patients with glioma. The clotting factor von Willebrand factor (VWF) is a highly adhesive procoagulant molecule that mediates platelet adhesion to endothelial and subendothelial surfaces. In the current analysis, we examined The Cancer Genome Atlas (TCGA) data to assess the effect of VWF gene expression on prognosis in patients with lower grade gliomas (LGGs). Methods: For newly diagnosed gliomas, we evaluated the association between and overall… Show more

“…Due to the significance of hemostasis, VWF plays a role in cancers by inhibiting angiogenesis and apoptosis (38)(39)(40). The high expression of VWF relates to endothelial damage and is used for indicating many pathological situations, including atherosclerosis, cardiovascular diseases, and cancers (41)(42)(43)(44). Besides, scientists pay attention to explore the relationship between the expression level of VWF and lung cancer.…”

“…29,30 Mutation in ATRX also had been reported, commonly in LGG. 31 In addition, we also found the mutations of these mutated genes in the patients with LGG were mainly missense mutations. A high proportion of missense mutations may change genetic information though affecting the structure and function of proteins, 32,33 suggesting possible roles in the pathogenesis of LGG.…”

“…Again, the preoperative antigen plasma median levels of vWF:Ag were significantly higher in GBM than in meningiomas and increased vWF levels were associated with a threefold higher risk of death in GBM patients (Marfia et al, 2016[ 39 ]). Even in lower grade gliomas, vWF gene expression had a negative prognostic effect as it emerged from the genomic data of The Cancer Genome Atlas (Lehrer et al, 2018[ 31 ]). There was a significant relationship between growing staining intensity of vWF in microvessels and increasing grade of glioma confirming that the most important pathologic criteria for the diagnosis of HGG is microvessel proliferation (Mahzouni et al, 2010[ 37 ]).…”
