Increased expression of ErbB‐2 in liver is associated with hepatitis B × antigen and shorter survival in patients with liver cancer

Abstract: Hepatitis B 3 antigen, or HBxAg, contributes importantly to the pathogenesis of hepatocellular carcinoma (HCC). Given that HBxAg constitutively activates b-catenin and that upregulated ErbB-2 promotes b-catenin signaling in other tumor types, experiments were designed to ask whether HBxAg was associated with upregulated expression of ErbB-2. When HBxAg positive and negative HepG2 cells were subjected to proteomics analysis, ErbB-2 was shown to be upregulated in HepG2X but not control cells. ErbB-2 was also str… Show more

“…HER-2 amplification has been reported in breast cancer, and a monoclonal antibody trastuzumab, has become an important therapeutic option for patients with HER-2 amplification (44). However, reports on the status of HER-2 in HCC are conflicting (45)(46)(47)(48). By FISH, we showed a high frequency of deletion for TP53.…”

“…Amplification of HER-2 was observed in 2 patients with higher CTC counts among the 11 patients analyzed. Such amplifications are uncommon in primary tumors of HCC (45,46), whereas some studies show frequent HER-2 overexpression in HCC (47,48). The frequency of HER-2 amplification of CTCs in HCC needs to be investigated with more patients so as to clarify whether it could be a candidate gene for targeted therapy in HCC.…”

“…In breast cancer, ErbB2 was found to upregulate b-catenin via inactivation of GSK-3b by the Erk pathway (13). Also, in hepatocellular carcinoma, upregulation of ErbB-2 expression increased the existence of b-catenin (13,27). Further, a study suggested that serum levels of ErbB2 might be associated with liver dysfunction (28).…”
