Increased expression of cytokines in liver and serum in patients with extrahepatic diseases

Abstract: To determine whether the liver plays an immunological role in certain extrahepatic disorders, we investigated the expression of interleukin (IL)-1 beta, IL-6, interferon (IFN)-gamma, and tumor necrosis factor (TNF)-alpha in 11 patients who had recovered from cholecystolithiasis, 12 patients with gastric cancer, 20 patients with chronic hepatitis, and 6 healthy controls. Cytokine mRNAs in the liver were detected by semiquantitative reverse transcribed-polymerase chain reaction. Serum cytokines and soluble IL-2 … Show more

“…Therefore, measurement of sIL-2R␣ seemed to be useful to detect the expression of IL-2R on activated T lymphocytes. Although values of serum sIL-2R␣ in glioma patients were not so high compared with cancer patients reported previously [19][20][21], serum sIL-2R␣ levels of patients with malignant glioma seemed to elevate with the worsening of performance status. In 4 of 16 patients with malignant gliomas, we could detect the increase of serum sIL-2R␣ concentrations compared with previous values before their MR images could show tumor recurrence.…”

“…Unstimulated human blood mononuclear cells from healthy volunteers secrete very low levels of cytokine proteins (e.g., IL-2 = 27 ± 22, IL-4 = 2 ± 4, IL-6 = 49 ± 56, IFN- γ = 12 ± 11, TNF- α = 15 ± 18 pg/ml in 16 donors, Sullivan et al, 2000). Similarly, low cytokine levels are detected in the serum of healthy human controls (e.g., IL-1 β = 9.4 ± 2.0, IL-6 = 16.0 ± 7.1 pg/ml, n = 6, Fukuma et al, 1996). While in vitro work has clearly demonstrated the sensitivity of the immune system to alcohol, the direction of change (up- or down-regulation) can depend on the cell type studied (e.g., Choi et al, 2005; mouse mixed glial cultures; Fernandez-Lizarbe et al, 2009; rat or mouse mixed glial cultures; Verma et al, 1993; human monocytes).…”

“…Possible mechanisms for bile duct stones and cholecystectomy are through inflammation and bile exposure. Gallstones are known to cause inflammation to adjacent organs [ 41 ], but gallbladder removal can alleviate inflammation associated with gallstones [ 42 ]. However, when the gallbladder is removed, the enterohepatic circulation of bile salts becomes continuous [ 43 ].…”
