Increased apoptotic cells in bone marrow biopsies from patients with aplastic anaemia

Abstract: In order to investigate the involvement of apoptosis in the pathogenesis of aplastic anaemia (AA) we determined the proportion of apoptotic cells in paraffin-embedded bone marrow biopsies from patients with aplastic anaemia using an in situ TdT-catalysed DNA nick end labelling (TUNEL) staining method. A significant increase in the proportion of mononuclear apoptotic cells was demonstrated in biopsies from patients with aplastic anaemia (8.19 +/- 1.45%) when compared with controls (2.07 +/- 0.86%). These data s… Show more

“…A recent report indicates a good correlation between apoptosis and p53 overexpression in the bone marrow of MDS patients [25]. Bone marrow samples in AA have shown increased apoptosis but a low number of proliferating cells compared with control samples [6,16,26]. Bone marrow samples from patients with AA in this study as well as in a previous study have consistently shown no overexpression of p53 [17].…”

“…A high apoptotic rate, however, is by no means unique to MDS and has been reported in other conditions including aplastic anemia (15), Fanconi's anemia (16), beta thalassemia (17), human immunodeficiency virus (HIV) myelopathy, and other inflammatory myelitis (18).…”

“…Bone marrow stem cell dynamics are predicted using a simpli"ed version of the model for tumor growth, with parameters for bone marrow stem cell kinetics replacing those for tumor cells: c"24 hr (Tannock, 1978;Baker et al, 1995); AI"2.0% (Callera & Falcao, 1997;Lee et al, 190 FIG. 2.…”
