Increase of Fatty Acid Oxidation and VLDL Assembly and Secretion Overexpression of PTEN in Cultured Hepatocytes of Newborn Calf

Abstract: Background: Phosphatase and tensin homolog (PTEN) is a potent tumor suppressor gene that also plays a vital role in regulating fatty acid metabolism. Here we attempted to elucidate the role of PTEN in the regulation of fatty acid oxidation and the assembly and secretion of very low density lipoprotein (VLDL) in dairy cow liver. Methods: We transfected primary culture calf hepatocytes with adenovirus-mediated PTEN overexpression vector (AD-GFP-PTEN).PTEN-overexpressing hepatocytes and control hepatocytes were o… Show more

“…This suggests that miR-2285f may inhibit leucine membrane transportation by down-regulating SLC7A8 to reduce the potential accumulation of leucine in the liver 58 . In addition to AA metabolism, bovine-specific miRNAs may regulate lipid and fatty acids metabolism in liver, which is also different from monogastric animals, especially in terms of the relatively lower-density lipoprotein in dairy cows 59 . Some of the bovine-specific miRNAs, such as miR-2285t (Higher expressed under RS, compared with CS) and miR-2419-5p (lower expressed under AL diet, compared with CS) in jejunum, were also involved in AA transport.…”

“…The virus titer ultimately reached 10 10 PFU/ml. The recombinant adenoviruses (AD-GFP- SREBP-1c) were used to infect hepatocytes, and the multiplicity of infections (MOI) were 25, 50, 100, 150, and 200 [9]. A fluorescence confocal microscope and flow cytometer were used to ensure the best AD-GFP-SREBP-1c concentration and the optimal time for GFP expression.…”

“…At the pathway level, the PTEN-AKT pathway is required for the initiation of lactation through the induction of autocrine prolactin (Chen et al, 2012 ). In addition, PTEN has been shown also to play a vital role in regulating fatty acid metabolism (Fu et al, 2012 ).…”
