Increase in fatty acids and flotillins upon resveratrol treatment of human breast cancer cells

Abstract: Flotillin-1 and flotillin-2 are highly conserved proteins that localize into cholesterol-rich microdomains in cellular membranes. Flotillins are closely related to the occurrence and development of various types of human cancers. Flotillin-1 is highly expressed in breast cancer, and the high expression level of flotillin-1 is significantly correlated with poorer patient survival. Here we studied the relationship between the formation of lipid rafts and the expression of flotillins and lipids in human breast ca… Show more

“…This result agrees with previous findings reporting resveratrol, curcumin and piperine inhibition of breast cancer cell proliferation in vitro. We have previously reported that resveratrol in association with melphalan triggered cytotoxic effects in MCF-7 and MDA-MB-231 cells in vitro [ 7 ] and increased fatty acids in MCF-7 and MDA-MB-231 cells [ 34 ]. Wang et al [ 35 ] demonstrated that resveratrol inhibits the growth of MDA-MB-231 cells in a dose-dependent manner as well as their solid lipid nanoparticle form, displaying IC 50 values of 72.06 and 40.82 µg/mL, respectively.…”

“…Flotillin and caveolin are considered as the peptide markers of the lipid rafts (Buschiazzo et al, 2013). The effect of treatment could affect the structure of the raft and modulate the cancer cells, for instance, resveratrol shifts flotillin from intracellular vesicles to the raft domains in plasmalemma (Gomes et al, 2019). Other research studies indicate that the reduction of plasma cholesterol content results in lipid raft destabilisation and dysfunction of the cells (Buschiazzo et al, 2013).…”

“…A possible explanation for our results showing that resveratrol had a more significant effect on lipid composition in MDA-MB-231 cells than in MCF-7 cells could be related to the higher concentration of resveratrol used in our study in MDA-MB-231 cells (200 μM) as compared to MCF-7 cells (80 μM). As recently shown by our group, these different concentrations were used to normalize the effects of resveratrol in cell viability in the two cell lines [18].…”

“…MCF-7 and MDA-MB-231 (2 × 10 5 ) cells were cultivated in 24-well plates, incubated with Resv and DMSO 0.1% (control) in serum-free DMEM medium supplemented with 32 Pi (1.5 mCi/mL) for 0, 2, 8, 24 h. en cells were subjected to lipid extraction as described above and the associated radioactivity was measured by liquid scintillation counting [17]. We have recently shown that concentrations of resveratrol above 50 μM induce a significant decrease (∼40-50%) in cell viability in MCF-7 cells, while the same concentration of resveratrol causes only a minor decrease (∼10-40%) in cell viability in MDA-MB-231 cells [18]. To normalize the effects of resveratrol in cell viability, we used 80 μM of resveratrol in MCF-7 cells and 200 μM of resveratrol in MDA-MB-231 cells.…”
