Thirty-five thousand people die as a result of more than 2.8 million antibiotic-resistant infections in the United States of America per year. Pseudomonas aeruginosa (P. aeruginosa) is classified a serious threat, the second-highest threat category of the U.S. Department of Health and Human Services. Among others, the World Health Organization (WHO) encourages the discovery and development of novel antibiotic classes with new targets and mechanisms of action without cross-resistance to existing classes. To find potential new target sites in pathogenic bacteria, such as P. aeruginosa, it is inevitable to fully understand the molecular mechanism of homeostasis, metabolism, regulation, growth, and resistances thereof. P. aeruginosa maintains a sophisticated copper defense cascade comprising three stages, resembling those of public safety organizations. These stages include copper scavenging, first responder, and second responder. Similar mechanisms are found in numerous pathogens. Here we compare the copper-dependent transcription regulators cueR and copRS of Escherichia coli (E. coli) and P. aeruginosa. Further, phylogenetic analysis and structural modelling of mexPQ-opmE reveal that this efflux pump is unlikely to be involved in the copper export of P. aeruginosa. Altogether, we present current understandings of the copper homeostasis in P. aeruginosa and potential new target sites for antimicrobial agents or a combinatorial drug regimen in the fight against multidrug resistant pathogens.

In response to the enormous threat to human survival and development caused by the large number of viruses, it is necessary to strengthen the defense against and elimination of viruses. Metallic materials have been used against viruses for thousands of years due to their broad-spectrum antiviral properties, wide sources and excellent physicochemical properties; in particular, metal nanoparticles have advanced biomedical research. However, researchers in different fields hold dissimilar views on the antiviral mechanisms, which has slowed down the antiviral application of metal nanoparticles. As such, this review begins with an exhaustive compilation of previously published work on the antiviral capacity of metal nanoparticles and other materials. Afterwards, the discussion is centered on the antiviral mechanisms of metal nanoparticles at the biological and physicochemical levels. Emphasis is placed on the fact that the strong reducibility of metal nanoparticles may be the main reason for their efficient inactivation of viruses. We hope that this review will benefit the promotion of metal nanoparticles in the antiviral field and expedite the construction of a barrier between humans and viruses.

The fate and transport of viruses in floatation systems is considerably important for accurate determination of the safety of reusing reclaimed water in the flotation process. Herein, simulation experiments on the floatation, adsorption and desorption were performed to examine the effect of initial virus concentration, pH and floatation reagents on the adsorption of viruses ΦΧ174 and MS2 onto copper–molybdenum ores. The transport of viruses in the flotation systems was also investigated. The viruses in the reclaimed water were rapidly adsorbed onto the ore particles, suggesting that tailing wastewater can be safely reused for floatation. However, the adsorbed viruses in the concentrates, middlings and tailings may pose health risks at certain exposure levels. The transport of viruses was dominated by their attachment to ore particles, with most being inactivated or irreversibly adsorbed. The removal and adsorption rates decreased as the initial virus concentration increased, and the removal rate decreased as pH was increased from 7.5 to 9.5. In comparison with MS2, ΦΧ174 was removed more effectively. This suggested that electrostatic repulsion is an important mechanism because MS2 has a greater negative charge. The attachment of both ΦΧ174 and MS2 onto the mineral particles increased significantly in the presence of PJ053 and CaO.
