Inactivation of Escherichia coli glycerol kinase by 5,5'-dithiobis(2-nitrobenzoic acid) and N-ethylmaleimide: evidence for nucleotide regulatory binding sites

Abstract: Glycerol kinase (EC 2.7.1.30, ATP:glycerol 3-phosphotransferase) from Escherichia coli is inactivated by 5,5'-dithiobis(2-nitrobenzoic acid) (DTNB) and by N-ethylmaleimide (NEM) in 0.1 M triethanolamine at pH 7 and 25 degrees C. The inactivation by DTNB is reversed by dithiothreitol. In the cases of both reagents, the kinetics of activity loss are pseudo first order. The dependencies of the rate constants on reagent concentration show that while the inactivation by NEM obeys second-order kinetics (k2app = 0.3 … Show more

“…The necessity of substrate binding indicates that the target proteins probably have to undergo a conformational change before unphosphorylated EIIA Glc can bind. Evidence for conformational changes associated with substrate binding has been presented for LacY (245,390), MelB (586), MalK (71,581), and GlpK (355,654,874).…”

“…Nevertheless, an interesting observation from the curves is that, in all cases, there was a tendency to saturation. This suggests that the inactivation velocity was dominated by the equilibrium between the association and dissociation of enzyme-compound complex [ 39 ], which means that, at high compound concentrations, the rate of inactivation will not change. This pattern was observed for TcTIM inactivators even using different types of molecules such as benzothiazole derivatives [ 17 ] or natural products [ 19 ].…”

“…H 2 production is inhibited completely by 0.5 mM N-ethylmaleimide (NEM) [50,51], an inhibitor of glycerol kinase [53], which catalyzes the formation of GP and then DHAP (see Fig. 2).…”
