Inactivated rabies vaccines: Standardization of an in vitro assay for residual viable virus detection

Abstract: Human rabies, a neglected viral zoonosis, is preventable through domestic animals vaccination and post-exposure prophylaxis using inactivated rabies vaccines. During vaccine production, several mandatory in vivo quality control trials, such as potency, live virus, and safety, are responsible for the use of large numbers of laboratory animals. Over the years, global organizations encouraged the development of alternative methods to reduce, replace and refine the use of animals in the pharmaceutical industry. In… Show more

“…Despite that, this technology remains the primary choice for managing many pathogens, including Influenza, SARS, MERS, etc., since other technologies, such as recombinant polypeptides, have not been able to successfully imitate the pathogen's epitope configuration. Thus, this classic strategy is efficiently used to produce vaccines for hepatitis A [83], flu viruses [84], and rabies [85]. Now, the same strategy is used to develop a vaccine for COVID-19.…”

“…Указанные особенности явились основанием для применения ОТ-ПЦР-РВ в производстве антирабических препаратов, в частности, для количественной оценки содержания антигена в материале для иммунизации продуцентов антирабической сыворотки 15 . Неоднократно отмечено использование ОТ-ПЦР-РВ в комплексе с методами иммунофлуоресценции для оценки полноты инактивации вируса при изготовлении вакцин [57][58][59].…”

“…Classical inactivated whole virus particle vaccines (WPVs) have been used for preventing seasonal influenza 21 and other infectious diseases [22][23][24][25] . In the production of inactivated vaccines, inactivation is a crucial step to ensure the vaccine is safe for use.…”
