Inactivated influenza virus, when presented on dendritic cells, elicits human CD8+ cytolytic T cell responses.

Abstract: SummaryInactivated or subunit virus preparations have been excellent vaccines for inducing antibody responses . Generation of cytolytic T cell responses, however, is thought to require replicating virus, primarily to provide sufficiently large amounts of cytoplasmic proteins for processing and presentation on major histocompatibility complex class I molecules by antigen-presenting cells. Potent human CD8+ cytolytic T cell responses to live replicating influenza A virus are generated when dendritic cells are us… Show more

“…Dendritic cells (DC) are potent APCs that, after infection, migrate from peripheral tissues to the lymph nodes and activate CD4 ϩ and CD8 ϩ T lymphocytes (13)(14)(15)(16). By using mutational analysis, this migration step has been shown to be crucial for viral pathogenesis (17).…”

“…DCs were then added in graded doses to constant numbers of purified T cells in 96-or 48-well plates (Costar, Cambridge, MA). After 7 days, the T cells were assayed for cytotoxic activity on a TAP Ϫ/Ϫ , HLA-A*0201 MHC class II-negative T2 cell line (American Type Culture Collection) pulsed with 1 M matrix peptide using a conventional 51 Cr release assay (22). Target cells were labeled with Na 51 CrO 4 as previously described (22).…”
