Waterlogging often occurs simultaneously with high temperatures during summer. The objective of this study was to characterize anaerobic metabolism and transcript abundance of fermentative enzymes in roots of Kentucky bluegrass (Poa pratensis L.) in response to short-term waterlogging and high temperature stresses. Grasses were subjected to four treatments: (1) well-drained under normal temperature (20/15°C, day/night; control); (2) waterlogging under normal temperature (WL); (3) well-drained under high temperature (35/30°C, day/night; HT); and (4) waterlogging under high temperature (WL + HT). Greater reductions in leaf elongation rate and shoot dry mass were observed under WL + HT than either stress alone. Root water-soluble carbohydrate concentration decreased 45% and 46% at 3 and 5 days of WL + HT, respectively, compared to the control. At 5 days, activities of root alcohol dehydrogenase increased 93% and 90% and lactate dehydrogenase increased 95% and 98% under WL and WL + HT, respectively, compared to the control. Transcript abundance of ADH and PDC genes in the roots were not or only slightly shown under the control or HT conditions but were induced by WL or WL + HT, particularly under WL. The results indicated that short-term WL + HT induced root anaerobic metabolism in a similar way to WL alone but had more severe effects on leaf growth and root WSC than did WL in Kentucky bluegrass.

This review focusses on Ca(2+)-mediated plant cell signaling and optical methods for in vivo [Ca2+] monitoring and imaging in plants. The cytosolic free calcium concentration has long been considered the central cellular key in plants. However, more and more data are turning up that critically question this view. Conflicting arguments show that there are still many open questions. One conclusion is that the cytosolic free Ca2+ concentration is just one of many cellular network parameters orchestrating complex cellular signaling. Novel experimental strategies which unveil interference of cellular parameters and communication of transduction pathways are required to understand this network. To date only optical methods are able to provide both kinetic and spatial information about cellular key parameters simultaneously. Focussing on calcium there are currently three classes of calcium indicators employed (i.e., chemical fluorescent dyes, luminescent indicators, and green-fluorescent-protein-based indicators). Properties and capabilities as well as advantages and disadvantages of these indicators when used in plant systems are discussed. Finally, general experimental strategies are mentioned which are able to answer open questions raised here.

Background and aims Plant adaptation to waterlogged conditions requires a set of morphological and physiological/biochemical changes. The formation of aerenchyma is one of the most crucial adaptive traits for waterlogging tolerance. Enzymatic scavenging may also potentially contribute to waterlogging tolerance by providing detoxification of reactive oxygen species (ROS). Methods Changes of root porosity (as an indicator of aerenchyma formation) and activities in leaves of four major antioxidant enzymes, γ-amino butyric acid (GABA) and lactic acid contents in roots were evaluated in six barley genotypes contrasting in waterlogging tolerance.Results Soil waterlogging caused significant increases in adventitious root porosity in all genotypes. Waterlogging-tolerant genotypes showed not only significantly higher adventitious root porosity than sensitive genotypes but also much faster development of aerenchyma. The greatest difference in adventitious root porosity among genotypes was observed after 7 days of Plant Soil waterlogging treatment. At the same time, antioxidant enzyme activities in leaves, GABA and lactic acid contents in roots did not correlate with waterlogging tolerance. Conclusions A faster formation of aerenchyma in adventitious roots is one of the key factors for waterlogging tolerance in barley. This protocol is recommended to be applied in future studies to identify molecular markers linked to this trait using appropriate mapping populations.
