In Vivo Evaluation of Magnetic Targeting in Mice Colon Tumors with Ultra-Magnetic Liposomes Monitored by MRI

Abstract: We have developed a method that allows a semi-quantitative evaluation of targeting efficiency in tumors, which could be applied to different T contrast agents.

“…Citrate-coated γ-Fe 2 O 3 (cit-Fe 2 O 3 ) MNPs used in this work was synthesized by the co-precipitation method as described in our previous studies [ 22 ] ( Figure 2 , Table S1 ). All the lipids were dissolved in CHCl 3 (Carlo Erba Reagents, Val-de-Reuil, France) at a concentration of 25 mg/mL.…”

“…Previously, we successfully prepared ultra-magnetic liposomes (UMLs) by a reverse phase evaporation method, which gave a higher MNPs loading efficiency (about 100 folds) compared to the classical thin film hydration method [ 20 , 21 ]. Thanks to high content of MNPs, these vehicles are able to give high T2 MRI contrast enhancement and targeting effect in vivo [ 22 ]. Based on these positive results, in this work, we ought to obtain magnetic lipoplexes that would benefit from targeting efficacy and MRI contrast enhancement.…”

“…Among them, magnetic liposomes, also called Ultra Magnetic Liposomes (UML) because they contain a high volume ratio of magnetic nanoparticles, have been developed [11] . Due to their magnetic charge which was previously determined by magnetization measurements around 20% in volume fraction, UML have shown the ability to be accumulated in xenografted murine colon tumors with a factor 3 increase as regard to non-targeted liposomes [12]. Despite the presence of magnetic nanoparticles there is enough free space to expect a high loading of CA4P in the aqueous part of the liposomes.…”

“…The normalized volume of the tumor was then calculated at each time point by dividing the volume of the tumor by one of the tumor at D0. To compare the efficiency of accumulation of the CA4P-UML depending on the treatment group, the previously described post-processing methodology %I0.25 was used [12]. Briefly, the pixel intensity distribution of each tumor was obtained from the T2-weighted images and the percentage of pixels under the value I0.25= 0.25x(Intensitymax-Intensitymin) was calculated and noted %I0.25.…”
