In Vivo Detection of Vascular Adhesion Protein-1 in Experimental Inflammation

Abstract: Vascular adhesion protein-1 (VAP-1) is an inflammation-inducible endothelial glycoprotein which mediates leukocyte-endothelial cell interactions. To study the pathogenetic significance of VAP-1 in inflammatory disorders, an in vivo immunodetection method was used to detect the regulation of luminally expressed VAP-1 in experimental skin and joint inflammation in the pig and dog. Moreover, VAP-1 was studied as a potential target to localize inflammation by radioimmunoscintigraphy. Up-regulation of VAP-1 in expe… Show more

“…However, a minor part of the signal is likely caused by the hepatic elimination of the antibody. Previously, Jaakkola et al studied the in vivo behavior of 123 I-labeled chimeric mouse-human anti-VAP-1 antibody in dogs and pigs using SPECT (11). Their results suggest that uptake in liver is VAP-1-specific.…”

“…VAP-1 has been shown to be inflammation inducible [12,26]. However, it remains unknown whether IRI induces VAP-1 enzymatic activity at the site of primary insult and in remote organs.…”

“…In endothelial cells, it is most abundant in high endothelial venules but also present in flat endothelial cells. In these cells, it is mainly present in intracellular vesicles, which then translocate the molecule onto the luminal surface upon certain inflammatory stimuli (7). VAP-1 belongs to the family of semicarbazide-sensitive amine oxidases (SSAOs), which catalyze oxidative deamination of primary amines in the reaction R-CH 2 -NH 2 + O 2 + H 2 O → R-CHO + NH 3 + H 2 O 2 .…”
