In Vivo Chemoembolization and Magnetic Resonance Imaging of Liver Tumors by Using Iron Oxide Nanoshell/Doxorubicin/Poly(vinyl alcohol) Hybrid Composites

Abstract: A hybrid composite made up of superparamagnetic iron oxide nanoshells encapsulating the anticancer drug doxorubicin and bound together by poly(vinyl alcohol) was developed. Transcatheter arterial delivery in an in vivo liver tumor model led to embolization of the liver tumor blood vessels. Embolization was followed by disassembly of the composite. The nanoshells were then able to pass through the leaky tumor vasculature into the tumor tissue, thereby leading to slow and sustained release of the drug. As well a… Show more

“…[10] Alternatively, in consideration of the essential role of glucose in providing energy for tumor metabolism, [11] we strategically starved the tumors by depleting the intratumoral glucose by a glucose-metabolic reaction, which oxidized glucose into gluconic acid and H 2 O 2 using the catalysis of glucose oxidase (GOx). [12] As we know, H 2 O 2 causes malignant transformation of normal cells at endogenous concentrations, [13] but in turn triggers cancerous cell death at high concentrations.…”

“…As an alternative solution, these nanocarriers could be modified with biofunctional molecules to improve their bioavailability as well as to facilitate the site‐specific accumulation. Some representative magnetism‐responsive drug delivery systems are shown in Table 47–51. In a typical study by Luo et al,47 the Fe 2 O 3 nanoparticles were coated with mesoporous silica and further functionalized with β‐CD modified polyethylenimine (PEI) on the surface.…”
