Canine oocytes require an extended period of culture (72 h) in vitro for nuclear maturation to the metaphase II stage, which also results in high degeneration. Canine cumulus oocyte complexes were isolated by slicing from ovaries collected after ovariohysterectomy and cultured in serum-free synthetic oviductal fluid incubated at low (5%) or high (20%) oxygen levels. Changes in oocyte nuclear maturation rates, H 2 O 2 levels within the oocytes and mRNAs of reactive oxygen species inhibitory genes superoxide dismutase 1 and 2 (SOD1 and 2), glutathione reductase (GSR), glutathione peroxidase (GPX1), and catalase (CAT) were quantified. Higher meiotic resumption from germinal vesicle breakdown up to MII was observed in low O 2 (41.8G13.1%) compared to high O 2 (15.8G8.2%) (PZ0.014) after 52 h of culture (nZ112). Extension of the culture period up to 84 h at low O 2 (nZ457 oocytes) produced the highest meiotic resumption at 72 h (64.1G6.0%; PZ0.008), compared with 52 h. Oocytes (nZ110) cultured in high O 2 contained higher levels of peroxidase measured using the 2 0 ,7 0 -dichlorodihydrofluorescein diacetate fluorescence assay after 72 h of culture compared with low O 2 (PZ0.004). High O 2 -cultured oocytes also showed higher amounts of SOD1, SOD2, GSR, GPX1, and CAT mRNA. Vitamin E in high oxygen level was able to decrease degeneration (PZ0.008) but had no improving effect on percentage of oocytes in MII. These results for the first time showed that low oxygen gas composition improves nuclear maturation rates and alleviates the oxidative stress for canine oocytes during in vitro maturation.

RESUMENLa gestación en la perra transcurre entre la fecundación y el parto, y su duración tiene importancia clínica. La compleja biología de los gametos y embriones influye en la duración de la gestación, cuyo lapso fisiológico fluctúa entre los 57 y 70 días. Este amplio rango puede estar influenciado por los métodos referenciales para la estimación del inicio de la gestación. El realizar montas o inseminaciones en base a detección de la ovulación permite estrechar el rango a 62 a 64 días. Una condición de alto impacto obstétrico es la gestación de un solo cachorro, la cual es más prolongada y representa alto riesgo de distocia. Dada la importancia clínica de una adecuada proyección de la duración de la gestación y de la predicción de la fecha de parto, se han desarrollado fórmulas matemá-ticas para el cálculo de la edad fetal y tiempo al parto a través de la sistematización de la ultrasonografía gestacional.Palabras clave: perra; duración de gestación; parto ABSTRACTThe pregnancy in the bitch occurs between fertilization and whelping, and its length has clinical importance. Chez la plupart des mammifères, les ovocytes sont bloqués en métaphase II au moment de l'ovulation et cette inhibition de la méiose est ensuite levée par la fécondation. Chez les chiennes et les autres femelles de canidés, les ovocytes sont libérés au stade de prophase I, et il faut encore attendre 48 à 72 heures pour qu'ils atteignent le stade de métaphase II et deviennent fécondables. Cette particularité constitue aujourd'hui un frein au développement des biotechnologies de la reproduction chez les canidés. En effet, dans les essais de maturation in vitro d'ovocytes canins, seuls 10 à 30 % des ovocytes atteignent le stade de métaphase au bout de 72 heures de culture. Chez la chienne, la maturation nucléaire se produisant dans l'oviducte, des substituts de l'oviducte (milieux de culture comme le Synthetic Oviductal Fluid, explants d'oviductes, cultures sur tapis de cellules tubaires) ont été utilisés pour les cultures d'ovocytes in vitro, afin d'en améliorer le rendement, mais sans grand succès jusqu'à maintenant. Cet échec peut être dû au manque de données sur la composition du liquide tubaire de la chienne. L'étude de ce microenvironnement prend donc tout son intérêt, celui-ci étant probablement assez différent de celui des autres femelles, ne serait-ce que par l'existence du processus de lutéinisation préovulatoire dans cette espèce. À terme, la conception d'un milieu de maturation sur la base de la composition du liquide tubaire pourrait être une voie intéressante pour augmenter les taux de maturation in vitro. Mots
