In vivo and in vitro safety evaluation of fermented Citrus sunki peel extract: acute and 90-day repeated oral toxicity studies with genotoxicity assessment

Abstract: Background Citrus sunki Hort. ex Tanaka peel has been traditionally used as an ingredient in folk medicine due to its therapeutic effects on promotion of splenic health and diuresis as well as relief of gastrointestinal symptoms. Although a growing interest in health-promoting natural products and the development of highly concentrated products have facilitated consumption of C. sunki peel, its safety assessment has not been explored, posing a potential health risk. In this study, we carried ou… Show more

“…The animals used in this study were obtained from Orient Bio Inc. (Gyeonggi-do, Korea) and housed in environment-controlled animal rooms (temperature at 20–26°C, relative humidity of 50 ± 20%, 12-h light/dark cycle, ventilation 10–15 times/hour, and light intensity 150–300 Lux) at the Association of Assessment and Accreditation for Laboratory Animal Care (AAALAC) International-accredited animal facility (#001160) in Seoul National University Hospital as previously described ( Park et al, 2020 ). Animals were given free access to a γ-irradiated rodent diet (Teklad certified irradiated global 18% protein rodent diet, 2918C, Envigo RSM Inc., IN, United States) and autoclaved water.…”

“…Specific pathogen-free Sprague–Dawley (SD) rats, ICR mice, and conventional beagle dogs were purchased from Orient bio Inc. (Gyeonggi-do, Korea) and housed in environmentally controlled animal rooms designed for each species at the AAALAC International-accredited animal facility (#001160) of Seoul National University Hospital as previously described ( Park et al, 2020b ). Briefly, each environmental parameter was maintained as follows: temperature at 20–26°C for rodents and 19–28°C for dogs, relative humidity of 50 ± 20%, 12-h light/dark cycle, ventilation 10–15 times/hour, and light intensity 150–300 Lux.…”

“…If API crosses the epithelial barrier, a battery of six assays of safety and activity that cannot be studied in humans should be included: mutagenicity, carcinogenicity, one (and two)‐generation reproduction toxicity, neurotoxicity, embryotoxicity, and genetic studies (Lard‐Whiteford et al, 2004). Mutagenicity and genetic studies evaluate chromosomal and structural DNA changes, such as duplications, insertions, or translocations (da Silva Dantas et al, 2020; Nakamura et al, 2020; Prado‐Ochoa et al, 2020), and identify gene mutations (Gao et al, 2020; Marcelino et al, 2020; Park et al, 2020). Carcinogenicity studies evaluate tumors' development after 18 in mice or hamsters or 24 months in rats (Dekant et al, 2020; Prado‐Ochoa et al, 2020; Saleh et al, 2020).…”
