In vitro toxicity of local anaesthetics and corticosteroids on supraspinatus tenocyte viability and metabolism

Abstract: SummaryBackground/ObjectiveThe purpose of this study was to evaluate supraspinatus tenocyte viability and metabolism in explants exposed to various local anaesthetics and corticosteroids. Our hypothesis was that the tendons exposed to these common injectates would have significantly decreased cell viability and metabolism compared with controls.MethodsSupraspinatus tendon explants were obtained from dogs, placed in a culture media, and randomly assigned to one of the following groups: culture media only (contr… Show more

“…However, the same steroid dose in uninjured tendons initiates a short-term response equivalent to that of structural injury. Nuelle et al 32 Y Peritendinous injection of lidocaine, betamethasone, and methylprednisolone results in significant supraspinatus tenotoxicity in vitro. Ghellioni et al 33 Y Y There is a dose-dependent reduction of mechanical resistance and histological modifications of rotator cuff tendon exposed to corticosteroids.…”

“…Since the above mentioned molecular probes almost cover the entire visible spectrum, to gate living cells in low cytometry one needs to use two corresponding dyes -7-AAD and Sytox Blue. The use of these dyes makes it possible to carry out a multiparametric analysis of the state of cells and, without any significant interference, to study up to 4 parameters simultaneously in the same cell [15]. Some of the listed parameters can be analyzed not only by flow cytometry, followed by quantitative data, but also by fluorescence microscopy.…”

“…During arthroscopic joint surgery, intra-articular administration of LAs is common to improve postoperative pain scores and reduce narcotic consumption [1,2]. LAs are also administered into the shoulder joint to treat rotator cuff injuries and diseases [5]. In addition, fat aspiration for a cosmetic purpose or isolation of autologous adipose-derived stem/progenitor cells (ADSCs) routinely involves an administration of LAs [6][7][8][9][10].…”
