A protocol for in vitro induction of tetraploids of the grape rootstock 101-14 Millardet et de Grasset (Vitis riparia 9 V. rupestris) (101-14 Mgt) (2n = 2x = 38) 9 Muscadinia rotundifolia cv. Trayshed (2n = 2x = 40) hybrids was established. Different explant materials were exposed to the antimitotic chemical agents colchicine and oryzalin, at three concentrations, for 24, 48 and 72 h. Treated in vitro shoot tips, anthers and pre-embryogenic cultures were regenerated, and their ploidy levels were confirmed by chromosome counts and flow cytometry. It is generally considered that anatomical characteristics of leaves, including leaf stomatal size and number of chloroplasts in guard cells, correlate with ploidy levels of grape plants. Chromosome counts and flow cytometry are labor intensive and difficult, while the number of chloroplasts in stomatal guard cells and stomatal size are easily measured and can be stable and reliable markers. Analysis of stomata revealed clear differences between regenerated diploid and tetraploid plants, and these measurements were used as a preliminary screen, followed by chromosome counts and flow cytometry for confirmation. This study presents the results obtained by various combinations of explants, antimitotic agents, exposure time, concentration and ploidy analysis to evaluate in vitro chromosome doubling. These results provide an easy and effective method for production of tetraploid, potentially fertile Vitis 9 Muscadinia hybrids for utilization in rootstock breeding programs.

In vitro conditions for Japanese quince polyploidisation were investigated. Microshoots and isolated cotyledons were treated with colchicine and oryzalin. Morphogenesis was more dependent on the concentration of colchicine or oryzalin than on the duration of exposure, genotype differences were observed. Low oryzalin concentrations had no impact on morphogenesis. Plants with changed chromosome numbers were obtained at 0.25-38 mM colchicine and 10-50 lM oryzalin concentrations. It was determined that stomata length is a suitable parameter for identifying putative Japanese quince tetraploids. Stomata of tetraploid shoots of the same clone were approximately 1/3 longer than in the diploids. It was shown that through polyploidisation gigas effect was not obtained in fruit size but tetraploids have reduced seed set and an increased proportion of fruit flesh.

Induction of Anthurium andraeanum “Arizona” tetraploid by colchicine in vitro

Tetraploids plants of Anthurium andraeanum ''Arizona'' were successfully induced after treating diploid tissue masses with colchicine. Masses originating from diploid aerial roots were treated with colchicine at three different concentrations (i.e., 0.1, 0.2, 0.3%) for about 3, 5 and 7 h, and then were transferred into Murashige and Skoog medium containing 3 mg/l BAP ? 0.2 mg/l 2,4-D. After 60 days, the survival rate and numbers of regenerative shoots were scored. The high concentration and longer duration sharply reduced survival rate. In contrast, the regeneration of plantlets was not noticeably affected by colchicine. Tetraploid plants were obtained in all treatments, but the percentage of induced tetraploids ranged from 0.2 to 7.6%. The best induction was obtained with a 5-h, treatment with 0.3% colchicine. The stomatal size of tetraploid plants was larger than in diploid plants; however, the stomatal density was lower than in diploid plants. Tetraploid plants possessed stronger petioles, thicker and deeper green leaves, and thicker and longer lived spathes in comparison with diploid plants. Abnormal spathes, such as double spathes or those lacking pedicels, were observed in tetraploid plants. Tetraploid plantlets could be regenerated via aerial roots; this technique could be applied to tetraploid plant propagation.
