In vitro synthesis of glycosaminoglycans in endocrine ophthalmopathy

Abstract: The effects of humoral and cell-mediated immunity on the glycosaminoglycan synthesis of retrobulbar fibroblasts was evaluated in patients with endocrine ophthalmopathy. After incubation with IgG and sera, secreted glycosaminoglycans, radiolabeled with D-6-3H-glucosamine and 35sulfate, were precipitated with cetylpyridinium chloride and ethanol. Hyaluronic acid synthesis of human retrobulbar fibroblasts after incubation with sera and IgG and after co-culture with lymphocytes was assessed by means of a radiometr… Show more

“…These did not correlate with the presence of TSH-R stimulating antibodies, but there was a close relationship between fibroblast stimulating activity and the severity of TAO (415). Attempts at finding specific antibodies in TAO that either bind to fibroblasts or stimulate GAG synthesis have been unsuccessful (416)(417)(418). Attempts at finding specific antibodies in TAO that either bind to fibroblasts or stimulate GAG synthesis have been unsuccessful (416)(417)(418).…”

“…Since alterations in GAG metabolism has been demonstrated in diabetes mellitus and other autoimmune endocrine diseases (Kahaly et al, 1990(Kahaly et al, , 1992a(Kahaly et al, ,b, 1994aSchuler et al, 1993, 1-Jansen et al, 1 996a, b, e), we hypothesized an increase of circulating GAG in IDDM-associated microangiopathy. Thus, the relationship between diabetic nephropathy, retinopathy, neuropathy and GAG as well as the influence of the duration of the disease, the metabolic state of IDDM patients and their late complications on the Patients were followed monthly (clinical investigation and metabolic control) in the university outpatient clinic.…”
