In vitro selection of RNA aptamers that inhibit the activity of type A botulinum neurotoxin

Abstract: The category A agent, botulinum neurotoxin (BoNT), is the most toxic molecule known to mankind. The endopeptidase activity of light chain domain of BoNT is the cause for the inhibition of the neurotransmitter release and the flaccid paralysis that leads to lethality in botulism. Currently, antidotes are not available to reverse the flaccid paralysis caused by BoNT. In the present study, we have identified three RNA aptamers through SELEX process, which bind strongly to the light chain of type A BoNT (BoNT/A) a… Show more

“…Optimized selection methods were used to minimize the time and amount of target required to obtain high affinity DNA aptamers for BoNT [50,51]. Inhibitory RNA aptamers that disrupt the toxicological mechanism of BoNT have been selected which may prove useful in the development of therapeutics [52]. DNA aptamers for BoNT have been developed into an aptamer beacon with a detection limit of 1 ng/mL [53].…”

“…Botulinum toxin is one of the most devastating toxin reported. DNA and RNA aptamers have been designed to inhibit the toxicity of Botulinum toxin [18][19][20].In various disease conditions, proteins play an important role as a mediator. α-Thrombin is involved in the thrombus formation by enzymatic cleavage of two sites of Prothrombin by activated Factor X (Xa) in the clotting process.…”

“…For decades, toxins became problematic to food safety and aptamer based techniques have been developed for the detection of toxin like Ochratoxin A with DNA aptamer [95]. Currently, Ochratoxin A has also been detected using electrochemical aptasensor [96], aflatoxins [97][98][99][100], neurotoxins [101] such as botulinum [102,103], antibiotics [104][105][106][107] as well as pesticides such as nitenpyram, chlorpyrifos and imidacloprid [108].…”
