In vitro secretion of zymogens by bovine pancreatic acini and ultra-structural analysis of exocytosis

Abstract: The aim of this study is to establish a bovine pancreatic acinar cell culture model with longer viability and functionality. The cells could be maintained in a functional state for upto 20 days with normal morphology. Cells were positive for amylase as observed by immunofluorescence staining. Acinar cells are spherical and range about 2–3 µm in diameter. The porosome formed by exocytosis and heterogenous enzyme granules of size ranging 100–300 nm were seen on the surface of cells by electron microscopy. The ac… Show more

“…The bovine pancreas is a rich source of many therapeutic enzymes [ 61 ]. The major function of the bovine pancreatic acinar cell is the synthesis, storage, and secretion of several digestive enzymes, such as proteases, amylase, lipase, elastase, and ribonucleases, to catalyze the food constituent hydrolysis into absorbable forms [ 30 ]. This is mostly applied in human therapeutic applications [ 28 , 29 , 31 , 62 ].…”

“…Many proteases from several origins are well studied for anti-microbial and anti-biofilm effects. Bovine pancreatic enzymes are an excellent source for the many therapeutic enzymes [ 28 , 29 , 30 , 31 ]. However, protease type I (PtI) from the bovine pancreas is still not evaluated for its potential against the biofilms.…”

“…Subsequently, smaller units of mature zymogen granules in the pancreas of N. natrix joined together to form larger ones, similar to mouse (Hammel, Lagunoff, & Galli, 2010). Although these zymogen granules exhibited ultrastructural features of mature granules, they probably still contain an inactive form of enzymes as described in bovines (Jayaveni, Nithyanandham, & Rose, 2016). Such granules were sporadically present near the lumen of acini in close proximity to the cell membrane in the embryonic pancreas of the grass snake.…”

“…PA cells were obtained as described [ 16 , 17 ]. Briefly, the procedure involved dairy bull calves pancreatic tissue followed by digesting it in a dissociation medium containing collagenase III (1 mg/ml) in Kreb-Ringer bicarbonate (KRB) [ 18 ] buffer with 5% BSA and incubated for 15 min with constant shaking until homogenous solution was obtained.…”
