In vitro replication and expression of hepatitis B virus from chronically infected primary chimpanzee hepatocytes

Abstract: Primary chimpanzee hepatocytes were maintained in vitro utilizing a serum-free medium. Hepatocyte functions were sustained throughout the culture period as demonstrated by the synthesis and secretion of liver-specific plasma proteins characteristic for differentiated hepatocytes. Hepatocyte cultures established from a chimpanzee chronically infected with human hepatitis B virus exhibited the synthesis and secretion of hepatitis B virus proteins into the medium. In addition, the de novo replication of hepatitis… Show more

“…The culture system used in this work was based on the one described previously for baboon and chimpanzee primary hepatocyte cultures Jacob et al, 1989). Slight modifications have been introduced, however, to take into account the specificity of human hepatocytes.…”

“…Hepatocytes were allowed to attach for 3 h in Williams medium E supplemented with 5% fetal bovine serum, at which time the cultures were washed and changed to a serum-free medium supplemented with growth factors and hormones (10). The cultivation and characterization of baboon and chimpanzee hepatocytes in serum-free medium have been described (7,8,10). Secretion of apolipoproteins Al and E by the hepatocyte cultures was monitored by Western immunoblot as stringent markers of differentiation.…”

“…In the studies described here, we set out to cultivate CHV in primary cultures of marmoset hepatocytes (10) because hepatocytes are one of the targets of CHV infection in vivo * Corresponding author. and in vitro cultures of these cells are known to be permissive for the replication of other hepatotropic viruses (7,8). Our initial goal was to demonstrate replication of CHV in cell culture by using CH-specific antisera from convalescent animals (17).…”

“…These animals were free of any HDV markers including HDV RNA and anti-HDAg antibodies in the serum and had no history of HDV infection. The procedures utilized for the isolation and culture of primary hepatocytes in a serumfree medium formulation have been described previously (15).…”
