In vitro quantification of melanoma tumor cell invasion

Abstract: In order to quantify the invasiveness of melanoma tumor cells in vitro, a modification of the amniotic basement membrane (BM) model, described by Liotta et al. (Cancer Letters, 11, 141, 1980), was used in combination with radiolabeled tumor cells. B16-F10 metastatic murine melanoma cells and a derived clone (B16-F10L) were prelabeled with 0.1 muCi/ml of [14C]thymidine for 20-24 h in serum-free medium at 37 degrees C. Following incubation, fetal bovine serum was added to a concentration of 5 per cent, and the c… Show more

“…In order for cells to penetrate the Matrigel, they must secrete proteolytic enzymes that digest the protein components of the matrix, thereby creating a route for migration. This technique has been modified into a miniature scale assay by Hendrix [31], but still necessitates the use of hundreds of microliters of reagents. The Boyden chamber and its modification to include Matrigel is poorly equipped to study changes in proliferation and morphology of migrating cells, but does model cell passage through biologically relevant structures that impede migrating neoplastic cells.…”

“…Invasiveness of tumor cells was determined using the Membrane Invasion Culture System [35,36]. Briefly, ceils (5 Ɨ 104 to 1 Ɨ 105 ) were radiolabeled with 0.25 ~Ci/ml [14C]thymidine or 0.30/iCi/ml [125I]deoxyuridine (NEN) for 48 h in medium containing 2% serum and then seeded into the upper compartment of the MICS chambers.…”

“…lnvasiveness of tumor cells was determined using the Membrane Invasion Culture System (MICS, Gehlsen et al, 1984;Gehlsen and Hendrix, 1986;Hendrix et al, 1985b) which is a modification of the amnion invasion assay described by Liotta et al (1980). Briefly, cells (5 ā€¢ 104 to 1 ā€¢ 105) were radiolabeled with 0.25 ~tCi/ml ~4C-thymidine or 0.30 ~tCi/ml [125I]deoxyuridine ([t25I]Udr), both from New England Nuclear (Boston, MA), for 48 h in medium containing 2% serum and then seeded into the upper compartment of the MICS chambers.…”
