An efficient and reproducible protocol for in vitro propagation of Dolichandra unguis-cati has been established for the first time from nodal segments. In order to enhance survival rate under ex vitro conditions, photosynthetic potential of in vitro grown plantlets was also studied through JIP test based analysis of polyphasic OJIP chlorophyll a fluorescence OJIP transients, density of active reaction centers, light harvesting efficiency, electron transfer rate, dissipation energy, maximum quantum yield of primary PSII photochemistry and photosynthetic performance index. The best morphogenetic in term of explants response (92.2 %), shoot number (3.43 AE 0.07) and shoot length (4.7 AE 0.31 cm) was obtained on Murashige and Skoog medium supplemented with 0.5 mg l À1 BAP and 1.0 mg l À1 TDZ. The shoots exhibited high frequency rhizogenesis on half strength medium augmented with 2.0 mg l À1 IAA. In vitro plantlets developed highest rate of photosynthesis on day 18 after the initiation of rhizogenesis. High survival rate (96.16%) under ex vitro conditions was observed when in vitro plantlets having high photosynthetic efficiency (Fv/Fm > 0.75) were subjected to hardening and acclimatization process. Plantlets with reduced photosynthetic performance exhibited low survival rate under natural conditions. The developed in vitro protocol will be useful for genetic improvement and multiplication of D. unguis-cati. The results of this study also show that photosynthetic screening of in vitro developed plantlets is highly essential after the rhizogenesis process to achieve higher survival rate under field conditions.

An efficient in vitro regeneration protocol through somatic embryogenesis was established from longitudinally bisected protocorm (lTCL) of an endangered orchid Dendrobium ovatum. The efficiency of EC and SEs from the protocorms significantly relied on the concentration of PGRs. MS medium supplemented with TDZ (1.0 mg/l) induced optimum of EC (31.8%) and SEs (28.1/explant). Similarly, ZEA (0.5 mg/l) induced EC (27.6%) and SEs (17.1/explant). The combined treatment of TDZ (1.0 mg/l) and NAA (0.5 mg/l) resulted in the maximum induction of EC (58.6%) and SEs (39.8/explant) in an upright incubated explant. In another combined effect of ZEA (1.0 mg/l) and NAA (0.5 mg/l) induced EC (43.8%) and SEs (24.3/explant), whereas the combination of BAP (0.5 mg/l) and NAA (0.5 mg/l) produced EC (34.4%) and SEs (16.8/explants). The induced EC and SEs where healthier on medium containing TDZ + NAA than on the medium containing ZEA+NAA and BAP+NAA. The orientation of lTCL explants seemed to interact with position to affect the morphogenesis. The lTCL explants incubated in the upright orientation induced higher percentage of EC and a greater number of SEs/explants (EC-58.6% and SEs-39.8/explants) than that in the inverted orientation (EC- 16.4% and SEs-9.1/explants) irrespective of the PGRs. The SEs developed from the EC and the intermediate stage of PLBs were finally differentiated into plantlets. About 89% of plantlets were successfully acclimatized in the green house conditions. The effects of nitric oxide (NO) donor sodium nitroprusside (SNP) and various growth regulators on callus induction and shoot organogenesis in the commercially and pharmaceutically important medicinal plant Valeriana jatamansi is presented. Four different growth regulators at different concentrations were tested for their callusing efficiency followed by supplementation of SNP to the best responding growth regulator. Maximum callusing frequency (91.18%) was observed with the supplementation of 1.5 mg/L 1-Naphthaleneacetic acid + 15 μM SNP. 10% Coconut water (CW) was found to be the best growth regulator for the induction of shoots. The multiple shoot formation was also found to be enhanced with the supplementation of SNP. Maximum response for in vitro shoot multiplication was observed when the medium was supplemented with 10% CW + 15 μM SNP (89.32%) as compared to shoots induced in medium supplemented with 1.5 mg/L Benzyl aminopurine + 15 μM SNP (79.46%) respectively. No separate root induction medium was required as CW and CW + SNP supplementation was found to be sufficient enough to produce profuse rooting of the in vitro regenerated plants. Results revealed that SNP enhanced the callus and shoot induction frequencies. Supplementation of SNP also significantly reduced callus browing, allowed the tissues to recover and regenerate. This proposed in vitro culture system will be an effective mass propagation strategy for both commercial utilization and conservation of this highly valued plant species. Key MessageFirst report using SNP and growth hormone supplementation for enhanced propagation in Valeriana jatamansi presented. Supplementation of SNP enhanced the callus, multiple shoot induction and reduced callus browning in the in vitro cultures of V. jatamansi.
