In vitro propagation of Sandersonia aurantiaca Hook using thidiazuron

Abstract: A reliable in vitro propagation system was developed for the cut flower crop sandersonia (Sandersonia aurantiaca Hook). Explants comprised immature embryos from developing seeds and meristems from sprouting tubers. MS media supplemented with a range of thidiazuron (TDZ) concentrations lead to the formation of multiple shoot clusters. To overcome the deleterious effect of longterm exposure to TDZ concentrations, a multistage protocol of 4-weekly cycles was used. This comprised exposure to a high TDZ (3.0 mg l -… Show more

“…However, this extremely high shoot proliferation rate was achieved with germinating zygotic embryos, which were recombinant genotypes, and the attributes of a mature tree originating from such material are difficult to judge. The effective use of TDZ as the sole growth regulating compound for adventitious shoot regeneration was reported in several plant species such as Sandersonia aurantiaca (Deroles et al 2010), Phyllanthus amarus (Nitnaware et al 2011), Ochna integerrima (Ma et al 2011a) and also in Viburnum dentatum where its use was supplemented with dark treatments (Dai et al 2011). In our experiments we found TDZ to be an inconvenient plant growth regulator which frequently induced the formation of fasciated shoots.…”

“…Thidizazuron (TDZ), a new synthetic cytokinin-like PGR, has been reported to induce high-frequency somatic embryogenesis and organogenesis in dicots, such as Petunia hybrida (Thirukkumaran et al 2009), Phaseolus vulgaris (Kwapata et al 2010), and Phyllanthus amarus (Nitnaware et al 2011). It has also been reported recently that TDZ is effective in inducing somatic embryogenesis and organogenesis in monocots (Shan et al 2000;Gari and Rashid 2004;Panaia et al 2004;Ganeshan et al 2006;Cheruvathur et al 2010;Deroles et al 2010). Other factors in addition to embryogenic callus induction and regeneration also affect transformation frequency, including Agrobacterium strain, antibiotics used for controlling agrobacteria growth, and plant regeneration from resistant calli.…”
