SummaryThe present study was conducted to analyse the chromosomes of blood culture and oocyte chromatin quality at the time of recovery in dromedary camels in relation to age. Twelve young (about one year) and twelve adult (4-10 years old) female camels were used. These animals with unknown reproductive history were slaughtered in Kerdasa abattoir (Giza province, Egypt). Blood samples were collected via sterile syringes from camels before slaughtering for chromosomal analysis. Oocytes from ovaries of both ages were aspirated from small antral follicles 1 to 5 mm in diameter and classified according to their quality into four categories. Nuclear status of cumulus oocytes complexes (COCs) were evaluated directly after collection. The results indicated that, the frequencies of chromosomal abnormalities and sister chromatid exchanges (SCE'S) were increased significantly (pϽ0.05) with age. An increase in structural aberrations could be observed. There were no significant differences between young and adult camels in total number and quality of oocytes. Statistically significant (pϽ0.01) differences were between percentages of germinal vesicle breakdown (GVBD) and Germinal vesicle (GV) in young and adult camels. It is concluded that, the increase of age may have significant effects on structural chromosomal aberrations, SCE'S and meiotic stages of immature camel oocytes but not on the number and quality of oocytes.

RESUMO -O uso de bezerras pré-púberes como doadoras de ovócitos oferece um considerável potencial para acelerar o ganho genético através da diminuição do intervalo de gerações, além de incrementar o aproveitamento de fêmeas bovinas de alto valor genético. Os objetivos deste estudo foram analisar o efeito da idade e do tratamento estimulatório com gonadotrofinas sobre a resposta folicular, a taxa de recuperação, a produção e a avaliação morfológica de ovócitos colhidos em bezerras da raça nelore (B. t. indicus) a partir de 10 meses de idade. As bezerras foram distribuídas aleatoriamente em três grupos (n=4) e receberam tratamentos hormonais distintos, repetidos nos mesmos animais nos três períodos, aos 10, 11 e 12 meses de idade. No dia zero (D0), todas as bezerras receberam um implante intravaginal de progesterona e, em D1, 2 mg de benzoato de estradiol intramuscular (im). Em D6, deu-se início à diferenciação dos tratamentos (T): T1-120 UI de FSH, divididos em quatro aplicações im a cada 12 horas; T2 -120 UI de FSH, em uma única aplicação subcutânea (sc), mais 300 UI de eCG sc, T3 -sem tratamento estimulatório. No D8, o implante de progesterona foi retirado e a punção folicular transvaginal, guiada por ultra-som (PFTV), foi realizada no D9. Dos 402 ovócitos coletados, 53 foram desnudados e medidos, incluindo a zona pelúcida, encontrando-se um diâmetro médio de 153,38 (8,06) µm. Não houve diferença significativa entre os tratamentos e entre as idades para quantidade de folículos ≥ 3 mm, de ovócitos recuperados e de ovócitos viáveis. A média de ovócitos recuperados e ovócitos viáveis para cada tratamento foi: T1-11,08 (6,20) e 59,58% (25,80), T2-12,75 (8,04) e 66,02% (36,71), T3 -9,66 (8,15) e 51,51% (30,69), respectivamente. O valor médio da taxa de recuperação foi de 78,70%. Materials and Methods Collection of ovaries and oocyte maturationOvaries were obtained from 21 pubertal heifers of Japanese Black and Holstein breeds through a spay device. The heifers in this experiment were 8 to 12 months of age and were housed in closed stalls. Transvaginal recovery of ovaries from heifers was performed without anesthesia. The spay device (Kimberling-rupp spay device; Jorgensen Labo. Inc., Colo, USA) was inserted into the vagina, pushed through the vaginal wall up to each ovary, and the ovaries were collected. For the control, ovaries from adult cows of the same breed, which were more than 2 years old, were obtained at a local abattoir. The collected ovaries were placed in physiological saline (0.85%(w/v) NaCl) at 25 to 30 C. The ovaries were then transported to the laboratory within 2 h. Follicular oocytes were collected by aspiration of follicles 1 to 7 mm in diameter, and the follicular contents were mixed with modified Dulbecco' s phosphate buffered saline (mPBS) (Embryoteck; Nippon Zenyaku Co., Koriyama, Japan). After collection, the oocytes were washed twice with mPBS and once with the maturation medium (25 mM-Hepes TCM-199 with Earle's salts; Gibco, Grand Island, NY, USA) supplemented with 5% fetal bovine serum (FBS) and 0.01 A/ml FSH (Denka, Kawasaki, Japan). Then the oocytes surrounded with sufficient cumulus cells, were pooled and the number of the cumulus oocyte complexes were recorded. All the oocytes were cultured in 2.5 ml of maturation medium for 21 to 22 h at 38.5 C in 5% CO 2 in air.
