In vitro plantlet regeneration of Schinopsis balansae (Anacardiaceae)

Abstract: A protocol for the in vitro regeneration of rooted plants from nodal single bud segments of 10-yearold Schinopsis balansae trees was developed. Nodal segments were harvested from actively growing shoots of plants grown from seeds and maintained in pots under greenhouse conditions, and from epicormic shoots obtained by forced flushing of branches. Culture of nodal segments on nutrient medium containing the mineral salts and vitamins of Murashige and Skoog medium at 1/4 strength (1/4 MS), supplemented with 100 m… Show more

“…The promotive role of BA for shoot differentiation has been documented in Cercis canadensis var. mexicana (Mackay et al 1995), Schinopsis balansae (Sansberro et al 2003), and Holarrhena antidysenterica (Kumar et al 2005). However, shoot growth was best on the basal medium and the lowest level of BA tested (Table 1, Fig.…”

“…Auxins are widely used for induction of roots on regenerated shoots. IBA was effective for development of shoots in various members of the Anarcardiaceae (Boggetti et al 1999;Sansberro et al 2003). These plantlets were transferred to small pots containing garden soil: vermiculite (1:1) and kept in a mist house for 14 d. After the development of new leaves, plants were moved to a greenhouse.…”

“…On the contrary, in the presence of PVP, the release of phenolics was increased resulting in reduction of sprouting frequency. Growth-enhancing effect of charcoal has been reported in tree species of Anacardiaceae in vitro, such as in Schinopsis balansae (Sansberro et al 2003) and cashewnut (Das et al 1996). The phenolics released by the tissues in subculture often inhibit the growth and morphogenesis (Onay 2003).…”

“…In the present study, the maximum promotive effect on percent bud-break and rate of shoot multiplication were observed with BA as compared to 2-iP or Kinetin. The promotive role of BA on shoot proliferation has been previously documented in Schinopsis balansae (Sansberro et al 2003), Holarrhena antidysenterica (Kumar et al 2005), Eclipta alba (Ray and Bhattacharya 2008), Nelumbo nucifera (Shou et al 2008), and Centaurea ultreiae (Mallón et al 2010). In the present work, 6.4-fold shoot multiplication was obtained in 64% cultures on hormone-free medium.…”
