In vitro intestinal permeability studies, pharmacokinetics and tissue distribution of 6-methylcoumarin after oral and intraperitoneal administration in Wistar rats

Abstract: 6-Methylcoumarin (6MC) is a semisynthetic coumarin with important in vitro and in vivo antiinflammatory activity. In order to continue the pre-clinical characterization of this molecule, in vitro intestinal permeability, plasma profile and tissue distribution after oral administration in rats were studied. The permeability of 6MC was evaluated by the Caco-2 cellular model in both the apical-basal (A-B) and basal-apical (B-A) directions. The pharmacokinetics and biodistribution were evaluated in rats after oral… Show more

“…The prevalence of these two different kinds of diseases could be because they are common in the study area, or medicinal plants are more useful and have potent activity against digestive problems. We may assert that traditional herbal therapies may play an influential role in treating digestive tract problems, as they are the target of valuable activity [ 30 ]. Some of the most important and commonly used plants included Withania coagulans (33 URs), Piper nigrum (33 URs), and Thymus linearis (32 URs), Olea europaea subsp.…”

“…Experiments were held in triplicates. The apparent permeability coefficient (P app ) of PIT solution and the optimized bilosomes was then calculated using equation (4) (Cárdenas et al, 2017 ). where: P app : Apparent permeability coefficient (cm/s) dc/dt: Cumulative concentration of drug (c) transported to the basolateral chamber as a function of time (t) and was estimated from the slope of the linear portion of the concentration vs. time plot ▪ A: Surface area of the monolayer filter C 0 : Initial concentration of drug in the apical chamber (μg/mL) …”

“…The average surface area of a rat’s intestine is approximately 1.25 cm 2 per cm [ 19 ], and the estimated average surface area of our everted gut sac model is 3.125 cm 2 . The apparent permeability coefficients (P app ) were calculated from Equation (3) [ 20 ]: …”
