In vitro induction of tumour-specific immunity V. Detection of common antigenic determinatnts of murine fibrosarcomas

Abstract: Summary.-Two 3-methylcholanthrene and a spontaneous BALB/c fibrosarcoma were examined for tumour-associated antigens (TAA) by in vivo and in vitro induction of tumour-immune responses. When BALB/c mice were immunized to these fibrosarcomas by surgical tumour removal, cross-reacting tumour-associated transplantation antigens (TATA) were detected on all 3 tumours. Cytotoxic effector cells (CL) were then induced in vitro by co-culture of BALB/c spleen cells with the spontaneous, or one of the carcinogen-induced f… Show more

“…The simple excision of growing tumor nodules, a procedure which elicits an effective transplantation immunity to TSTA, was insufficient to generate detectable anti-tumor PEC in our assay. Restimulation of such primed PEC by in v i m homologous tumor cells proved to be difficult due to the release of lymphocytotoxic substances by fibrosarcoma cells and to the appearance of strong cross-reacting reactions which may obscure the specific lysis against unique determinants (Burton and Warner, 1978).…”

“…In addition to the unique antigens, the cold inhibition experiment revealed a possible presence of weak common antigens which have been detected on similar tumors by both in vivo and in vitro studies and suggested to be either virus-related or fetal determinants (Burton and Warner, 1978;Hellstrom et al, 1978).…”

“…Tumor rejection assays , however, have almost invariably not identified common cross-reacting TATA. Several exceptions among chemically-induced neoplasms have been reported (Leffell and Coggin, 1977;Economu et al 1977;Burton and Warner, 1978;Fritze et al, 1976). Hellstrom et al (1979) have described both in vitro and in vivo crossreactivity among some chemically-induced BALB/c sarcomas; the in vivo-detected cross-reactivity, indicating the presence of common or shared TATA, was stated to be strongest against those sarcomas expressing murine leukemia virus (MuLV)-associated antigens; these are found on cell surfaces or within many transplantable neoplasms of the mouse.…”
