To evaluate mesometrial transplantation of frozen-thawed ovarian tissue in rabbit and to choose the optimized fertilization method for oocytes retrieved from grafts by investigating the capability of oocyte fertilization and further development. Forty rabbits were divided into three groups randomly: control group, fresh tissues transplantation group and frozen-thawed tissues transplantation group. Three months after the transplantation, rabbits were stimulated with FSH and oocytes were retrieved 13 h after human chorionic gonadotropin (HCG) injection. Oocytes matured in vivo or in vitro were then fertilized by conventional in vitro fertilization (IVF) or intracytoplasmic sperm injection (ICSI), followed by observation and evaluation of fertilization rate and blastocyst formation rate. Blastocytes embryos were transferred to pseudopregnancy rabbits to observe pregnancy rate and birth rate. There were no significant differences in the percentage of oocytes matured either in vivo or in vitro among the three groups. The fertilization rate, cleavage rate and blastocyst formation rate of in vivo-matured oocytes had no difference among the three groups, whether they were fertilized by IVF or ICSI. Significantly higher fertilization rates of in vitro-matured oocytes were observed with ICSI compared with IVF in each group. The blastocyst formation rate of in vitro-matured oocytes was significantly lower than that of in vivo-matured oocytes in each group. The birth rate of in vivo-matured oocytes was significantly higher than that of in vitro-matured oocytes, although the pregnancy rate was similar between them. Mesometrial transplantation of frozen-thawed ovarian tissue may provide favorable conditions for follicle development. Oocytes retrieved from mesometrial grafts can develop to the blastocyst stage and produce live offspring. ICSI can optimize the fertilization rate of in vitro-matured oocytes retrieved from grafts.

Rabbits are both productive and classic laboratory animals. Some particularities of female reproductive physiology make the rabbit an extraordinary model for the study of embryology and assisted reproductive techniques. For instance, as the ovulation is induced, the embryo development can be known with accuracy. Embryos are surrounded by a mucin coat which is crucial to prevent embryo mortality. Besides, the anatomy of the uterus does not allow embryo transmigration between both uterine horns, and so it is possible to test different reproductive techniques. Knowledge on early embryo development, and on influencing factors, has allowed to develop new insights into embryo manipulation, such as recovery, transfer, cryopreservation, in vitro fertilisation, cloning, or transgenesis. Also the rabbit may be used as a model for human reproductive health, because rabbit embryo and feto-placental development are similar to the human. This chapter reviews the aspects of the reproductive physiology in the female rabbit and discusses some embryo manipulation techniques available in the species.

Özet: Bu çalışmada iki farklı süperovülatör ajanın tavşan oositlerinin kalite, kantite ve in vitro fertilizasyonu üzerine etkisini araştırmak amaçlanmıştır. Araştırmada 16 dişi, 2 erkek Beyaz Yeni Zelanda ırkı tavşan kullanılmıştır. Bir grup dişi tavşana (n=8) kas içi yolla 24 saat arayla 3 kez 75 IU PMSG, diğer bir grup tavşana (n=8) ise kas içi yolla günde 2 kez 3 gün süreyle 7.5 IU FSH enjekte edilmiştir. Son hormon uygulamasından 24 saat sonra her iki gruba 100 IU hCG damar içi yolla enjekte edilmiştir. HCG enjeksiyonundan 15 saat sonra oviduktlar yıkanarak oositler toplanmıştır. PMSG uygulanan grupta toplam 34 adet oosit elde edilirken, FSH uygulanan grupta toplam 77 oosit alınmıştır. Gruplar arasındaki oosit sayısı farkı önemli bulunmuştur (p<0.01). Dejenere oosit oranı PMSG uygulanan grupta %14.71 (5/34), FSH uygulanan grupta ise %18.18 (14/77) olarak kaydedilmiştir. Ancak, gruplar arası dejenere oosit sayısı farkının istatistiki açıdan önemli olmadığı saptanmıştır (p>0.05). Oositlerin in vitro fertilizasyonunun değerlendiril-mesinde, fertilizasyona alınan oositlerin ikiye bölünme aşamasına ulaşmaları kriter alınmıştır. Kültür periyodu sonunda fertilizasyon oranları PMSG uygulanan grupta % 44.83 (13/29), FSH uygulanan grupta ise %52.38 (33/63) bulunmuştur. Farkların istatistiki açı-dan önemli olmamasına rağmen kantitatif farklılık tavşanlarda süperovulasyon oluşturmak için FSH kullanımını desteklemektedir (p>0.05). Ancak, tavşanlarda FSH ile daha etkili süperovulasyonun uygulanabilmesine rağmen, daha ucuz oluşu ve uygulama kolaylığı gibi avantajlarından dolayı PMSG'nin de tercih edilebileceği kanısına varılmıştır.Anahtar sözcükler: In vitro fertilizasyon , oosit, süperovulasyon, tavşan.. Effect of different superovulator agents on the oocyte quality and in vitro fertilization in rabbitsSummary: The aim of this study was to investigate the effect of two different superovulator agents on oocyte quality, quantity and in vitro fertilization of rabbits. In this study 16 female and 2 male New Zealand White rabbits were used. A group of female rabbits (n=8) was intramusculary injected with 75 IU PMSG 3 times with 24 hours intervals. Another group of female rabbits (n=8) was injected (intramuscular) with 7.5 IU FSH twice daily during three consecutive days. Twentyfour hours after the last hormon injection both groups of animals ovulation was induced by an intravenous injection of 100 IU hCG. Fifteen hours after hCG injection, oocytes were collected by flushing the oviducts. A total of 34 oocytes were obtained from PMSG applied group and 77 oocytes were collected from FSH applied group. Difference between groups for oocyte number was found significant (p<0.01). In the PMSG applied group the percentage of degenerated oocytes was 14.71 % (5/34), and in the FSH applied group it was 18.18 % (14/77). The difference between groups for degenerated oocytes was not statistically significant (p>0.05).
