In Vitro Differentiation of Mouse Granulocytes

Abstract: Granulocytes are central players of the immune system and, once activated, a tightly controlled balance between effector functions and cell removal by apoptosis guarantees maximal host benefit with least possible collateral damage to healthy tissue.Granulocytes are terminally differentiated cells that cannot be maintained in culture for prolonged times. Isolating primary granulocytes is inefficient and challenging when working with mice, and especially so for the lowly abundant eosinophil and basophil subtypes… Show more

“…We performed qPCR array analysis in IL-3 cond Hoxb8 basophils originating from conditionally immortalized mouse bone marrow progenitors 27 , 28 (further referred to as in vitro -differentiated mouse basophils) to study the impact of IL-3 on genes regulating apoptosis. As shown in Figure 1 , IL-3 increased pro-survival genes such as Bcl-2 , Bcl-x L and Mcl-1 in mouse basophils, while pro-apoptotic genes like caspase-2, -3, -9 and Bim were downregulated (see Supplementary Table S1 for full array list).…”

“…Using in vitro -differentiated mouse basophils, 27 , 28 we revealed that BCL-2 and BCL-X L prevent apoptotic cell death at steady state and identified both BCL-2 and BCL-X L as potential targets to counteract the IL-3-mediated pro-survival effects. Especially the combined inhibition of both proteins by ABT-263 was able to fully neutralize the IL-3 induced survival advantage.…”

Whether these cells can serve as a reliable experimental tool to support or replace animal studies will depend on whether they can fully recapitulate the functions of terminally differentiated neutrophils.…”

“…The technique offers advantages over other myeloid cell lines due to the non-cancerous origin of cells and their physiological differentiation programme. Although the technique has been further developed by other groups (Gurzeler et al 2013;Reinhart et al 2016), no studies have been published so far using these cells to investigate SOCE. Whether these cells can serve as a reliable experimental tool to support or replace animal studies will depend on whether they can fully recapitulate the functions of terminally differentiated neutrophils.…”
