In Vitro Comparison of Chen Medium and Optisol-GS Medium for Human Corneal Storage

Abstract: Corneas stored in CM were thicker during storage than those stored in OM. The two storage media did not differ with respect to endothelial cell loss during storage or to the percentage of TUNEL-positive cells or keratocyte density at the end of the storage period.

“…In a toxicity trial of Optisol-GS, which contains GM at a concentration of 100 μg/ml, SEM studies of human corneas after 7 days of storage in the solution detected no toxicity. 27 In our study, a 200 μg/ml dose of GM resulted in some endothelial cell damage, although not severe. It may be of great benefi t if new corneal storage media could be developed that cause less damage to endothelial cells than do currently available products.…”

“…Optisol-GS is a storage medium normally used to preserve postmortem human donor corneal tissue for the purpose of transplantation 31,32. It maintains epithelial, stromal, and endothelial viability for up to 14 days after corneal extraction 31,32.…”

“…Nelson and associates compared Chen medium with Optisol-GS in their study. 8 Although corneas stored in Chen medium were found thicker during storage than those stored in Optisol-GS medium, they reported no difference between 2 media regarding endothelial cell loss or keratocyte density during storage.…”
