In Vitro Autoradiographic Visualization of Guanosine‐5’‐O‐(3‐[35S]Thio)triphosphate Binding Stimulated by Sphingosine 1‐Phosphate and Lysophosphatidic Acid

Abstract: S]GTP␥S binding in the olfactory bulb, glia limitans, and cortical subventricular zone of 1-day-old rats, whereas enhanced labeling was not observed in the latter area of 5-day-old rats. Sphingosine 1-phosphate stimulated binding in the cortical and striatal subventricular zones and olfactory bulb in 1-and 5-day-old rats. In the absence of radioligand for sphingosine 1-phosphate and lysophosphatidic acid receptors, [ 35 S]GTP␥S autoradiography provides a unique opportunity to study the spatial distribution, on… Show more

“…Treatment with NEM therefore reveals if members of the G i/o class of G proteins are involved in certain signalling processes. In [ 35 S]GTPgS autoradiography studies, both basal signal and agonist-induced responses are sensitive to NEM, this having been observed for several receptors (Waeber and Chiu, 1999;Laitinen, 2004;Palomäki and Laitinen, 2006). In the present study, pre-treatment with NEM (1 mM) for 30 min clearly decreased the overall labelling observed under basal conditions (Fig.…”

“…35 S]GTP␥S autoradiography using S1P has revealed S1P receptor-mediated G-protein activity throughout the brain (Waeber and Chiu, 1999). Of interest in the present study is the apparent overlap in the distribution of S1P and CB 1 receptor-mediated activity in regions including the cerebellum, hippocampus, striatum, and cortex.…”

“…CB 2 receptors are located primarily in the immune system, whereas CB 1 receptors are abundant in the CNS and are present in several peripheral organs (Howlett et al, 2002). CB 1 and S1P receptors stimulate G-protein activity in the CNS and are codistributed in many regions (Sim et al, 1995;Waeber and Chiu, 1999). Interest in the S1P receptors has increased because of the therapeutic potential of the immunomodulatory drug FTY720, which targets all of these receptors, except S1P 2 .…”

“…Using S1P-induced 35 S-GTP␥S labeling as a measure of receptor localization, different brain regions have quite variable levels of expression (Sim-Selley et al 2009;Waeber and Chiu 1999). For example, labeling in the cerebellum is high, whereas that in the thalamus is relatively low.…”
