Methanol leaf extract of Eucalyptus camaldulensis was investigated for phytochemical compositions, antioxidants, antimicrobial and safety profile. The antibacterial study was carried out using agar well diffusion method, while antioxidant activities were evaluated by 2, 2′diphenyl-1-picrylhydrazyl (DPPH) and ferric reducing antioxidant power (FRAP) assays. A total of fifteen rats were divided into three groups (5 rats each) and were given 0, 250 and 500 mg/kg bwt of the extract orally for 28 days. Results revealed that tannins (24.72±0.36 g) is the most abundant phytochemicals followed by phenols (6.01±0.89 mg/g) while alkaloid (0.19±0.67 mg/g) was the least. Extract demonstrated antioxidant activities with IC 50 of the 244.98±5.24 µg/mL and 462.755 ± 6.98 µg/mL in DPPH and FRAP assays respectively. The extract inhibited the bacteria growth with minimum inhibitory concentrations (MIC) and minimal bactericidal concentrations (MBC) values ranged between 7.5-60 μg mL -1 and 60-12 μg mL -1 respectively . The concentrations of albumins, bilirubins sodium, potassium, creatinine, serum transaminases and alkaline phosphatase (ALP) activities were not significantly (p>0.05) altered by the extract. Urea concentration was significantly (p<0.05) higher while proteins were lower in rats treated with 500 mg/kg bw of the extract. Methanol extract of E. camaldulensis could be considered as a cheap source of effective and safe herbal remedy with potential candidate for the development of a new drug.

Acute and sub-acute (30-day) toxicity studies of Aegialitis rotundifolia Roxb., leaves extract in Wistar rats: safety assessment of a rare mangrove traditionally utilized as pain antidote Abstract Background: Aegialitis rotundifolia Roxb., (Plumbaginaceae) is a small mangrove tree or shrub traditionally used by the local healers of mangroves for curing pain and inflammation. However, there are no scientific reports of its toxicological properties which would guarantee the safety of its folkloric usage as a potent pain reliever. Therefore, the present study deals in acute and sub-acute (30-day) toxicity studies of Aegialitis rotundifolia ethanolic leaves extract (ARELE) on male and female Wistar rats administered through oral gavage.Methods: Acute toxicity study was conducted at a single oral dose of 1000, 1500, and 2000 mg/kg, body weight (b.wt.) for 14 days with a special emphasis on the first four hours after drug administration to find out any mortality and morbidity. In sub-acute toxicity, the extract at the doses of 125, 250 and 500 mg/kg, b.wt., was administered orally for 30 days. Important parameters such as general behaviour, body and organ weight, urinalysis, haematological and biochemical profile, organ macroscopy and microscopy were conducted. Organ sample of liver, kidney, pancreas, heart, lungs, and stomach were taken from both male and female rats, whereas the sample of testis and ovary was taken from male and female rats respectively for organ necropsy and histopathological studies. Neurobehavioral toxicity was conducted by performing a functional observational battery (FOB) and locomotor activity on the initial and final week of the 30-day study period. Results:No mortality or any major signs of morbidity was recorded for acute toxicity except for the limit dose (2000 mg/kg, b.wt.) which produced a slight short-term sedative effect. In sub-acute toxicity, no major alteration was observed in the evaluated parameters. However, few minor changes were recorded for high dose (500 mg/kg, b.wt.) group. Conclusion:The results of the present study showed that oral administration of Aegialitis rotundifolia ethanolic leaves extract did not produce any severe toxic effects in both acute and sub-acute studies in Wistar rats. Therefore, usage of an appropriate amount of Aegialitis rotundifolia leaves extract preferably at low doses for its traditional use should be considered safe.

Background: Zingiber officinalis known as ginger, under the family Zingiberaceae, is a popular food spice and occupies an important place in Ayurvedic and traditional medicine. Its rhizome is used as antioxidative, carminative and for chronic diseases. Aims: Study indicated the curative effects of methanolic extract of ginger on severe exercise-induced oxidative damage in vital organs. Materials and Methods: Oxidative stress was established in male rat by forced swimming in an acrylic plastic pool filled with water. The total duration of 8 h per day including rest was allowed for both the treated and untreated forced swimming group of animals. Oxidative stress has been observed by monitoring of SOD, CAT, GST and Px activities along with quantification of TBARS and CD levels in liver, cerebrum, testis, and plasma. Whether the applied dose has toxicity if any, general body weight parameter was assessed. Results: Results indicate that SOD, CAT, GST and Px activities were decreased along with increase in levels of TBARS and CD in aforesaid tissues significantly in forced swimming group which were corrected significantly after 15 days pre-treatment followed by 28 days co-treatment of methanolic extract at the concentration of 20 mg/0.5 ml olive oil/100 gm body weight/rat/day in forced swimming group of rats (p<0.05). The extract showed non-toxic effect reflected here from the study of percentage gain in body weight. Conclusion: This finding reveals that methanolic extract of rhizome of Zingiber officinalis exhibited significant protective and curative effects on vital tissues in swimming-induced oxidative stressed rats.
