The purposes of this study were to evaluate the effectiveness of 2% chlorhexidine (CHX) gluconate gel, calcium hydroxide [Ca(OH) 2 ] and their combination with iodoform and zinc oxide powder as intracanal medications against select microorganisms, and to measure the pH changes caused by these medications. Antimicrobial activity was determined by the agar diffusion method. The zones of growth inhibition were measured and the results were analyzed statistically by Kruskal-Wallis test (p<0.05). The pH of the pastes was measured right after preparation, after 24 h and 1 week later. The largest mean zones of microbial inhibition were produced by 2% CHX gel, followed by Ca(OH) 2 + 2% CHX gel + iodoform, Ca(OH) 2 + 2% CHX gel, Ca(OH) 2 + 2% CHX gel + zinc oxide, and Ca(OH) 2 + water. The mean pH of all medications stayed above 12.0 during the whole experiment, except for CHX gel (pH=7.0). The results of this study showed that all medications had antimicrobial activity, but the most effective against the tested microorganisms were 2% CHX gel, followed by its combination with Ca(OH) 2 and iodoform.

Candida albicans is present in the oral cavity and in the whole digestive tract of humans and other animals, being frequently related to endodontic treatment failure. The present study determined the incidence of C. albicans in the oral cavity and the susceptibility of isolates to different pH values and saturated calcium hydroxide aqueous solution at pH 12.5. Sixty-five patients attending the Endodontic Clinic at the Sagrado Coração University participated in the study. The collected samples were cultivated in selective media for C. albicans and the isolates were tested in terms of resistance to both alkaline pH and saturated aqueous solution of calcium hydroxide. In relation to time variables, yeast viability was assessed by the Sabouraud's agar culture and fluorescein diacetate and ethidium bromide fluorescent staining method. Results from the different pHs and experimental times, including those from different techniques measuring fungal viability, were compared using the chi-square and Fisher's exact tests (α=0.05). The yeasts became completely inviable after 48 h of contact with the calcium hydroxide solution. On the other hand, when exposed to the alkaline culture broth, the yeasts were found to be viable at pHs 9.5 and 10.5 for up to 7 days. In conclusion, C. albicans can only be completely inhibited by direct contact with saturated calcium hydroxide aqueous solution after 48 h of exposure.

Resumo. Na presente pesquisa, cujo objetivo foi avaliar a ação antimicrobiana in vitro do extrato de própolis verde (também denominado propólis neste estudo) e de associações medicamentosas com e sem essa substância contra o microrganismo Enterococcus faecalis, utilizou-se a técnica de difusão em ágar pelo método do poço, e a cepa indicadora Enterococcus faecalis ATCC 29212. A avaliação do experimento, que se deu pela medição do halo de inibição do crescimento microbiano, contemplou dez grupos: G1 -soro fisiológico; G2 -pasta de hidróxido de cálcio pró-análise + soro fisiológico; G3 -paramonoclorofenol canforado (PMCC); G4 -Rifocort R ; G5 -pasta de Guedes-Pinto (iodofórmio + PMCC + Rifocort R ); G6 -própolis; G7 -própolis + pasta de hidróxido de cálcio pró-análise; G8 -própolis + Rifocort R + iodofórmio; G9 -própolis + Rifocort R ; G10 -própolis + iodofórmio. A solução salina e o hidróxido de cálcio + soro fisiológico não apresentaram atividade antimicrobiana. Já o Rifocort R , o extrato de própolis verde + Rifocort R , a pasta Guedes-Pinto e o extrato de própolis verde + iodofórmio + Rifocort R mostraram-se mais efetivos contra o Enterococcus faecalis do que as outras substâncias, apresentando halo de inibição de 10,5mm; 9,3mm, 9mm e 8,5, respectivamente, com diferença estatisticamente significante (p<0,05). Os resultados obtidos possibilitaram as seguintes conclusões: o hidróxido de cálcio e a pasta de iodofórmio não apresentaram ação antimicrobiana; o melhor resultado foi proporcionado pelo Rifocort R ; a solução de extrato de própolis verde apresentou baixa atividade antimicrobiana contra o E. faecalis. In the present research whose objective was to evaluate the in vitro antimicrobial activity of propolis extract (also called propolis in this study) and combination therapy with and without the substance against the microorganism Enterococcus faecalis, we used diffusion technique in agar by pit method, and the indicator strain Enterococcus faecalis ATCC 29212. The evaluation of the experiment, which took place by measuring the inhibition of microbial growth halo, looked ten groups: G1 -saline; G2 -calcium hydroxide slurry for analysis + saline; G3 -paramonochlorophenol camphor (ECCP); G4 -Rifocort R ; G5 -Guedes-Pinto paste (iodoform + Rifocort R PMCC); G6 -propolis; G7 -propolis + calcium hydroxide paste for analysis; G8 -propolis + Rifocort R + iodoform; G9 -propolis + Rifocort R ; G10 -propolis + iodoform. The salt solution and calcium hydroxide + saline showed no antimicrobial activity. Already Rifocort R , green propolis extract + Rifocort R , Guedes-Pinto paste and green propolis extract + iodoform + Rifocort R were more effective against Enterococcus faecalis than the other substances, with inhibition halo 10.5mm; 9,3mm, 9mm and 8.5, respectively, with a statistically significant difference (p <0.05).
