In vitro andin vivo impact of a new glycosphingolipid on neutrophils

Abstract: A new water-soluble, orally absorbable de-N-acetyl-lysoganglioside (WILD20), breakdown product of the monosialoganglioside GM1, was found to influence some parameters of neutrophil response to inflammation stimuli. Superoxide anion production appears inhibited, along with neutrophil killing properties. A block of both pathways of arachidonic acid cascade and PAF was also found, as well as neutrophil ICAM-1-mediated adhesion to endothelial cells. Of particular interest was the significant reduction of neutrophi… Show more

“…To substantiate these findings, release of AA, thromboxane, LTs, PAF, and other pro-inflammatory eicosanoids was observed in a variety of cells, in in vitro and in vivo settings [144,145]. Additionally, Wild-20 reduced human platelet aggregation, phorbol ester-and f-MLP-induced O2 -production in human PMNs, ICAM-mediated neutrophil adhesion, and cell invasion in a murine peritonitis model [144,145]. What seems curious is the observation that Wild-20 strongly inhibited porcine pPLA2 (at 10 -9 M) but not sPLA2 and cPLA2 in corresponding enzyme assays [144,145].…”

“…Although Wild-20 failed directly to inhibit PKC, it reduced PKC translocation to the membrane, and as a consequence cPLA2 activity was greatly diminished since cPLA2 has to be phosphorylated and translocated to the membrane [19,96].
