In vitro and in vivo activities of a new quinolone, WIN 57273, possessing potent activity against gram-positive bacteria

Abstract: The antibacterial activity of a new 7-dimethylpyridinyl quinolone, WIN 57273, was assessed by using in vitro and in vivo models. Agar inclusion and broth dilution in vitro tests revealed broad-spectrum activity against gram-positive and selected gram-negative organisms, with the greatest potency observed against the staphylococci. The MIC for 90% of coagulase-positive strains tested (MIC90) was <0.002 ,ug/ml; for the coagulasenegative strains the MIC90 was 0.008 ,ug/ml. Against enterococci the MIC90 was 0.06 ,… Show more

“…Inflammation and the achievement of a steady state in serum both appear to increase drug penetration into the vitreous humor (23). New investigational quinolones have excellent activity against the gram-positive pathogens (19,26,32,35) that are most commonly isolated in bacterial endophthalmitis (11,25,29). These observations, in conjunction with results of our pharmacokinetic studies, suggest that the fluoroquinolone antimicrobial agents may have a role in the prophylaxis and therapy of bacterial endophthalmitis.…”

“…Win 57273 is a new broad-spectrum 7-dimethylpyridinyl quinolone with greater activity against gram-positive organisms than the activity of ciprofloxacin or ofloxacin; the compound is also active against Legionella species (10,19,20,31). Pharmacokinetic data in rats revealed maximum VOL.…”

“…Additionally, the widespread use of ciprofloxacin has resulted in resistance to this quinolone among gentamicin-resistant enterococci (16). A number of novel fluoroquinolones, including WIN 57273 (6,7,17), PD (10) were performed in triplicate with each strain to determine the extents and rates of killing by WIN 57273 alone or with penicillin and by penicillin with and without an aminoglycoside. Antibiotics were tested at the following concentrations: penicillin, 10 U/ml; WIN 57273, 2 jig/ml; gentamicin, 5 ,ug/ml; and streptomycin, 25 p,g/ml.…”
