In vitro and in vivo evaluation of two carrageenan-based formulations to prevent HPV acquisition

Abstract: Commercial vaccines against human papillomavirus (HPV) have low uptake due to parental autonomy, dosing regimen, cost, and cold chain storage requirements. Carrageenan (CG)-based formulations prevent HPV infection in vitro and in vivo but data are needed on the durability of anti-HPV activity and the effect of seminal plasma (SP). The Population Council's PC-515 gel and the lubricant Divine 9 were tested for their physicochemical properties and anti-HPV activity against HPV16, 18, and 45 pseudoviruses (PsVs). … Show more

“…In support of earlier data demonstrating that anti-HIV-1 (cellbased assays), anti-SHIV-RT (macaque vaginal explants), and anti-HSV-2 (murine model) activity of PC-1005 and anti-HPV activity (murine model) of CG is maintained in the presence of biological fluids (SP) (14,20,22,23;Kenney et al,unpublished), PC-1005 was active against HIV-1 BaL and HSV-2 in human ectocervical explants in the presence of vaginal fluids. Infection levels were inversely correlated with MIV-150 levels (HIV-1) and CG (HSV-2) levels in the CVLs.…”

“…Kenney Zydowsky, and N. Teleshova, submitted for publication), and its activity is not affected by SP (20). Furthermore, PC-1005 gel provides CG-mediated activity against HPV, and this activity is not compromised in the presence of SP (22,23).…”

“…On the contrary, the MZC gel or MZCL IVR (Table 1) combines carrageenan (CG) and zinc (in addition to MIV-150, an anti-HIV non-nucleoside RTI) showed potent and durable synergistic anti-HSV-2 activity [20-22]. This MPT formulation retains antiviral activities when virus is added in seminal plasma [23]. …”

“…CG, however, was by far the most potent, with an IC 50 of 2 ng/ml, and it was active against a broad spectrum of genital HPV types. Importantly, it retains high activity in the presence of seminal plasma [23]. Interestingly, several commercial sexual lubricants are formulated using carrageenan as the primary gelling agent.…”

“…Consequently, heparin and other polyanionic compounds have been reported to act as HSPG antagonists, binding and sequestering HPV in the extracel-lular environment and thus hampering the cell surface attachment of HPV and hence infection (references 11 and 12 and references therein). The in vivo effectiveness of this anti-HPV strategy was demonstrated recently using the polyanionic sugar carrageenan (13,14).…”
