In the Immune Synapse, ZAP-70 Controls T Cell Polarization and Recruitment of Signaling Proteins but Not Formation of the Synaptic Pattern

Abstract: Recognition by T cells of their ligands at the surface of antigen-presenting cells (APCs) leads to T cell activation, polarization of the T cell toward the APC, and formation of an immune synapse. Using ZAP-70-deficient T cells expressing zeta-GFP, we show that ZAP-70 signaling drives the TCR-dependent reorientation of the microtubule-organizing center thus leading to relocation of a zeta-GFP(+) intracellular compartment close to the APC. ZAP-70 is also necessary to supply the synapse with the signaling molecu… Show more

“…5F). To verify that accumulation of clathrin, actin, dynamin-2, Arp2/3 complex, CD2AP and Hip1R at the IS was specific and to validate the Synapsis Measures software, we analyzed the cytoplasmic accumulation of GFP and CD45, a membrane tyrosine phosphatase, which after initial cell-cell contact does not accumulate at the IS (Blanchard et al, 2002;Freiberg et al, 2002;Varma et al, 2006), in T-cell-APC conjugates in the presence and absence of antigen. Endogenous CD45 and GFP did not accumulate at the IS after antigen stimulation in control or clathrin KD T cells (supplementary material Fig.…”

“…In T cells, ZAP-70 is known to be required for clustering signaling molecules into lipid rafts. 20 We assumed that the expression of ZAP-70 in a subset of CLL patients might also effect the activation of signaling molecules associated with the B-cell receptor.…”

“…CD45 (ϩ3.7) is tyrosine phosphatase required for lymphocyte activation. Zap70 (T cell receptor -chain associated protein kinase, 70 kDa, ϩ3.1) plays a vital signaling role in the interaction between antigen presenting cells and T cells that leads to T cell activation (19). T cell factor 1 (ϩ8.4) is critical for T cell maturation (20), whereas B lymphoid kinase (ϩ2.2) is important for B cell proliferation and developmental progression (21).…”
