In situ hybridization of mRNA for β-preprotachykinin and preprosomatostatin in adult rat dorsal root ganglia: comparison with immunocytochemical localization

Abstract: In situ hybridization histochemistry was used to identify neurons in rat dorsal root ganglia that contained mRNAs encoding beta-preprotachykinin and preprosomatostatin. The distribution of these neurons was compared with the distribution of neurons containing tachykinins or somatostatin, identified using immunocytochemical techniques. Neurons labelled for beta-preprotachykinin mRNA constituted 20% of the total neuronal population and belonged to the small cell class. Neurons labelled for preprosomatostatin mRN… Show more

“…A similar qual- (Leonard et al, 1988;Oblinger et al, 1989;Wong and Oblinger, 1990) have been used effectively in semiquantitative studies, and it might be useful to employ these probes in size frequency histogram analyses such as that shown in Figure 4A for NF-L and peripherin. Such size frequency histogram analyses, using in situ hybridization histochemistry , have been done successfully for various neuropeptides and alphatubulin in the rat DRG (Henken et a]., 1988;Boehmer et al, 1989). However, our data (Table 1B) indicate that the neuropeptides, substance P and CGRP, cut across the cell size boundaries defined by the neurofilament and peripherin antigenicities.…”

“…2 in Szaro et al, 1990). Furthermore, in situ hybridization analysis of NF mRNA reveals its presence in the small cell perikarya, although at very low levels (Hoffman et al, 1987;Wong and Oblinger, 1987;Verge et al, 1990). The reverse situation is the case for peripherin.…”

“…The IncStar antiserum crossreacts with several tachykinins (Henken, Tessler, Chesselet, Hudson, Baldino & Murray, 1988). The Peninsula antiserum is claimed by the vendor to have less than 1 % cross-reactivity with either neurokinin A or B.…”

“…Previous reports have shown that SOM-LI and SOM-mRNA are present in only a small number of primary afferents. In rats, approximately 10% of all dorsal root ganglion cells were immunopositive Molander et al, 1987;Henken et al, 1988) or showed a positive reaction after in situ hybridization (Henken et al, 1988). All these studies were performed without colchicine treatment of the ganglia.…”
