AbstractLoss of biodiversity and altered ecosystem functioning are driven by the cumulative effects of multiple natural and anthropogenic stressors affecting both quantity and quality of water resources. Here we performed a 40-day laboratory microcosm experiment to assess the individual and combined effects of drought and the model fungicide tebuconazole (TBZ) on leaf litter decomposition (LLD), a fundamental biogeochemical process in freshwater ecosystems. Starting out from a worst-case scenario perspective, leaf-associated microbial communities were exposed to severe drought conditions (four 5-day drought periods alternated with 4-day immersion periods) and/or a chronic exposure to TBZ (nominal concentration of 20 µg L−1). We assessed the direct effects of drought and fungicide on the structure (biomass, diversity) and activity (extracellular enzymatic potential) of fungal and bacterial assemblages colonizing leaves. We also investigated indirect effects on the feeding rates of the amphipod Gammarus fossarum on leaves previously exposed to drought and/or TBZ contamination. Results indicate a stronger effect of drought stress than fungicide contamination under the experimental conditions applied. Indeed, the drought stress strongly impacted microbial community structure and activities, inhibiting the LLD process and leading to cascading effects on macroinvertebrate feeding. However, despite the lack of significant effect of TBZ applied alone, the effects of drought on microbial functions (i.e., decrease in LLD and in enzymatic activities) and on Gammarus feeding rates were more pronounced when drought and TBZ stresses were applied together. In a perspective of ecological risk assessment and ecosystem management for sustainability, these findings stress the need for deeper insight into how multiple stressors can affect the functioning of aquatic ecosystems and associated services.

Deciphering evolutionary processes occurring within long-term contaminated wild populations is essential for the ecological risk assessment of persistent chemical contaminations. Using field populations of Gammarus, a commonly-used genus in aquatic ecotoxicology, the present study sought to gain insights into the extent to which long-term exposure to metals in the field could effectively lead to shifts in toxicological sensitivities. For this, we identified a Gammarus population inhabiting a stream contaminated by cadmium (Cd). We compared the Cd-exposure and Cd-sensitivity of this population to those of five reference populations. Active biomonitoring determined in different years and seasons that significant levels of Cd were bioavailable in the contaminated site. Laboratory sensitivity tests under common garden conditions established that this long-term field exposure led to the development of a moderate Cd tolerance, which was maintained after a 3-week acclimatization in the laboratory, and transmitted to offspring produced under clean conditions. The potential physiological costs of tolerance were assessed by means of feeding rate measurements (in the laboratory and in situ). They revealed that, unlike for reference populations, the feeding activity of organisms from the tolerant population was greatly decreased when they were maintained under laboratory conditions, potentially indicating a high population vulnerability to environmental perturbations. Because dissolved Cd concentrations in water from the contaminated site were low (averaging 0.045 µg L(-1)) and below the current European environmental quality standard for Cd for inland surface waters (fixed at 0.08 µg L(-1) in soft water environments), this case study sheds light onto the extent to which current environmental quality standards are protective against potential adverse outcomes of adaptive and micro-evolutionary processes occurring in contaminated environments.

Gammarids are important members of a stream's macrozoobenthos biocoenosis and food web. Moreover, they proved to be very sensitive towards different types of pollution. GamTox TM is a new in situ ecotoxicity test, based on survival and feeding behavior of caged gammarids for active monitoring of small streams in agricultural areas. GamTox TM has been applied in two streams with specific pollution problems in the catchment of Lake Constance. Ten organisms were exposed in 5 replicates in flow through test tubes containing one conditioned alder leaf, placed in baskets which were attached in the stream bottom and on the banks. Each week, the number of living animals was counted, the percentage of leaf skeletized estimated in semi-quantitative classes and a new elder leaf provided. Dead organisms were removed. Simultaneously, chemical analyses of pesticides and nutrients (N-compounds, P) were performed on cumulative water samples over one week. Moreover, macrozoobenthos was collected and determined according to the IBCH method, and the SPEAR index calculated. GamTox TM proved to be very sensitive to detect pesticides, copper as well as nutrients, both during acute pollution pulses and chronic exposures of up to 6 weeks. Survival turned out to be a more sensitive and less variable parameter than feeding. GamTox TM is easy to perform and directly provides a measure of ecotoxicological effects of toxicant/nutrient mixtures, which cannot be predicted by biological indices based on macrozoobenthos data such as IBCH and SPEAR-index.
