In Silico Structure-Guided Optimization and Molecular Simulation Studies of 3-Phenoxy-4-(3-trifluoromethylphenyl)pyridazines as Potent Phytoene Desaturase Inhibitors

Abstract: A series of novel 3-phenoxy-4-(3-trifluoromethylphenyl)pyridazines 2–5 were designed, based on the structure of our previous lead compound 1 through the in silico structure-guided optimization approach. The results showed that some of these new compounds showed a good herbicidal activity at the rate of 750 g ai/ha by both pre- and post-emergence applications, especially compound 2a, which displayed a comparable pre-emergence herbicidal activity to diflufenican at 300–750 g ai/ha, and a higher post-emergence he… Show more

“…Three days after application, the stems of wheat and rice began to show albinism, resulting in severe chemical injury. It was initially speculated that compounds ( Rac )- 6q and ( S )- 6q contained triazole functional groups, which had strong internal absorption, and wheat roots were multicut and developed, which were important causes of pesticide injury. , At the same time, compounds ( Rac )-BF, ( S )-BF, ( Rac )- 6h , and ( S )- 6h did not injure wheat, which also verified that ( Rac )-BF and ( S )-BF were mainly used for weeding wheat fields . Therefore, compounds ( Rac )-BF, ( Rac )- 6h , and ( Rac )- 6q had higher selectivity than the S configuration for six crops.…”

“…But weed infestation poses a significant threat to crop yields, as they compete with crops for nutrients, light, and water, leading to yield reductions of more than one-third. Effective weed control through herbicide application is crucial to increasing crop productivity. However, prolonged and extensive use of single herbicides, such as acetyi coa-carboxylase (ACCase) and acetolactate synthase (ALS) inhibitors, has resulted in severe weed resistance . This necessitates higher herbicide dosages to eradicate resistant weeds, resulting in significant environmental pollution. Therefore, to overcome weed resistance, there is an urgent need to develop novel herbicides with high activity, high selectivity, and environmental friendliness …”

“…The overexpression and purification methods were similar to those in our previous reports . BL21(DE3) chemically competent cells transformed with plasmid DNA (pET-15b- Synechococcus PDS) were cultured in 500 mL of 2×YT medium containing 100 μg/mL ampicillin until the cell culture density was 0.6 at OD 600 .…”
