In silico binding affinity to cyclooxygenase-II and green synthesis of benzylpyrazolyl coumarin derivatives

Abstract: Here, we have reported enhanced molecular binding affinities of benzylpyrazolyl coumarin scaffolds with COX-II inhibitor and an ionic liquid catalyzed simple green synthetic route to access these scaffolds.

“…Thus, [C 4 MIM][BF 4 ]–water mixtures allowed effective dispersion of hydrophobic C 60 fullerenes, which showed high biomedical potential . Benzylpyrazolyl coumarin scaffolds, which presumably possessed inhibitory activity toward the cyclooxygenase COX-II, were synthesized by using [C 5 MIM][Br] as catalyst and reaction media . The enzyme phytase self-assembled into active nanospheres in [C 4 MIM][BF 4 ], and these nanospheres were subsequently used for production of platinum-decorated, curcumin-loaded phytase nanospeheres, which the authors considered “multifunctional protein-based drug delivery vehicles” …”

“…The more amino acid residues that interact with the ligands, the smaller the value of ΔG will be (negative) and the reaction will take place spontaneously and bind very strongly so that the inhibitory effect it creates will also be stronger [13] .…”

“…The use of ionic liquid makes this approach environmentally and ecofriendly benign (Scheme 11b). [59] Another alternative approach for the synthesis of a vast array of pyrazolyl-coumarin derivatives 49 has been established by Jadhav and Shloorkar in the same year (Scheme 11c). [60] By using 10 mol% of alum [KAl(SO 4 ) 2 • 12H 2 O] as the transitionmetal-free catalyst, the desired products 49 derived from the multicomponent reaction of 4-hydroxycoumarin 1, aldehydes 31, hydrazines 47, and ethyl acetoacetate 48 under microwave irradiation, has been obtained in 70-97% yield.…”
