In-depth proteomic profiling of the uveal melanoma secretome

Abstract: Uveal melanoma (UM), the most common primary intraocular tumour in adults, is characterised by a high frequency of metastases to the liver, typically with a fatal outcome. Proteins secreted from cancer cells (‘secretome’) are biologically important molecules thought to contribute to tumour progression. We examined the UM secretome by applying a label-free nanoLCMS/MS proteomic approach to profile proteins secreted into culture media by primary UM tumours with a high− (HR; n = 11) or low− (LR; n = 4) metastatic… Show more

“…In contrast, a cluster of biomarkers for one disease would be a better diagnostic tool with much higher sensitivity, specificity, and clinical accuracy. Therefore, new investigations called "proteomic profiling" or "multimarker profiling" focus on the identification of multiple co-expressed biomarkers or signature biomarker patterns which allow early detection, staging, therapeutic monitoring, and prognostic predictions (7,(84)(85)(86)(87)(88). This approach can be adopted for both serum and tissue specimens.…”

“…Combined flow-throughs were acidified with Trifluoroacetic acid to 0.2% (v/v). Incubation media was bound to and digested off Strataclean beads as previously described (Angi et al, 2016;Ashraf Kharaz et al, 2017). A yeast enolase peptide standard was added to each sample, though this was not utilised for later quantification.…”
