In vitro culture of ginger is highly influenced by medium formulations such as sucrose and growth regulator. A micro-propagation was done to determine the influence of sucrose concentration and growth regulator on in vitro growth of red ginger. Sterilized sprouts with shoot tips collected from the rhizome were cultured on formulated media of MS medium containing three diverse concentrations of sucrose and different types and combinations of growth regulators for shoot differentiation. The number of days required for buds to break varies from 13 - 36 according to the sucrose concentration, type, and combinations of growth regulators. Among the formulated medium, the MS medium contained 30 gr of sucrose and BA 1 mg.l-1 induced bud sprouts in about 13-14 days. The sucrose concentration and growth regulator also had significant differences in shoot number, leaf number, and root number. Interaction of BA 1 mg.l-1 and sucrose 30 g.l-1 produced the highest number of shoots per explant (6.33) and the number of leaves per shoot (3.72). Auxin promotes the growth of intact roots as the medium supplemented with NAA yielded a higher number of roots, however, rooting was also observed in all formulated media after two passages of subculturing.

Berginia ligulata is medicinally significant and endangered plant of the western Himalayan Region. This plant is used to treat many diseases such as urinary problems and heart problems. By using leaf explant, an efficient and rapid micropropagation protocol was developed. In in-vitro propagation, MS medium was supplemented with benzyl amino purine (BAP), 1-naphthalene acetic acid (NAA) and indol-3-acetic acid (IAA) to induce multiple shoots from leaf explants. The most positive response for the formation of calluses was observed on MS media supplemented with MS medium + BAP(1 mg/L) + NAA(1 mg/L) followed by MS medium + BAP (0.5 mg/L) + NAA (1 mg/L) after 3 weeks. Maximum number of shoots were obtained on MS medium + BAP (2 mg/L) +IAA (2 mg/L). Maximum % shoot response was obtained on MS medium + Kn (2.5 mg/L) concentration. The maximum number of roots was observed on MS medium without growth regulators. These well-rooted plants were established in pots and acclimatized in the lab and transferred to field conditions. This process is useful to produce endangered plants on large scale.

Pelargonium Radula as a Plant Bioindicator in Monitoring Mercury in Drinking Water

Pollution by organic and inorganic toxic substances has raised public and environmental concern globally since last decade. However, due to rapid growth of world population along with excessive industrial development, this situation worsens. One of the non-essential heavy metals, mercury (Hg), is a widespread toxic pollutant commonly found in drinking water sources. Therefore, there is a need to monitor the level of Hg in the drinking water. Biomonitoring, which use biological response to assess environmental changes, is one of the approaches that are getting more attention recently. Plant bioindicator offers huge advantages over conventional water quality analysis. Other than cheap, easy to apply and give rapid results; most importantly, people in the rural area can apply this method to monitor water quality without the need of modern equipment and technical expertise. In this study, young leaves from terrestrial plants were treated with water containing Hg solutions. Plant that showed visible morphological changes was selected as potential bioindicator and further analyzed. Over 60 plants were screened including herbs, shrubs and flowering plants. At the 24th h of observation, 15 plants showed morphological changes with several obvious symptoms, including the presence of dark spot, chlorosis, browning of leaves and wrinkle. Among these plants, Plectranthus amboinicus, Lantana camara and Pelargonium radula treated with Hg solutions exhibited morphological changes at the 6th h of treatment, compared to the control. In the present study, Pelargonium radula was chosen as the Hg bioindicator as it gave the fastest and visible morphological changes, which is within 4 h of treatment. This new finding was promising, as it demonstrated that plants could be used for Hg biomonitoring for the safety of drinking water.
